CaM plays a key role in many cellular processes. In the CNS, it is involved in synaptic transmission and neuronal plasticity associated with short-term and longterm potentiation, and learning and memory processes.

It is well known that signal transduction of pain initiates in primary afferent neurons and transmits to the dorsal horn of the spinal cord (Scholz and Woolf, 2002). Signal transduction in the spinal cord is important for nociception, but dynamic change (i.e., central sensitization) and neuronal plasticity associated with glial activation also have important roles in neuropathic pain (Inoue et al., 2007 Tsuda et al., 2005). A comparison of the DCS value estimated using in silica, HTS and rat in situ perfusion estimated for a given subset of clinical candidates is given in Table 1 based on internal historical data. As indicated, in silica data predicted DCS class correctly in 74% of cases and HTS data in 81% of cases using rat in situ perfusion as the reference data set. In many cases, errors were associated with the misclassification of DCS Class I or III with class II or IV (permeability errors). Misclassifications based on solubility errors were less common. Also some very poorly soluble Class II compounds can masquerade as Class IV candidates based on precipitation and adsorption to the hlters and plastics associated with the permeability apparatus. 

The resins in the friction dust area tend to be rigid and the flexibility and plasticity associated with the long alkyl chain of phenolic lipids have been used in natural rubber vulcanisation by for example incorporating crosslinking with phosphorylated cardanol (ref. 252). Unpolymerised CNSL phenols have been used in natural or diene rubber compositions for tyre treads to give an improved dynamic elastic modulus but with the same hardness as formulations without the phenolic addition (ref. 253).

Historically, some plasticizers associated with plastic pipes contained lead and leaching of lead into drinking water was found to be possible. Such plasticizers are not now used and while the problem is detectable it is not associated with high level lead leaching.
