The concept of inhibition via p elimination of fluoride ion has now been extended to the irreversible inhibition of a-amino acid decarboxylases. Ornithine decarboxylase (ODC), which catalyzes the decarboxylation of ornithine to putrescine is irreversibly inhibited by a-difluoromethylornithine (IX Fig. 9) (28). In this case, the carbanion formation which precedes P elimination is generated by loss of CO2, and not by proton abstraction (Fig. 9). Similarly, aromatic amino acid decarboxylase is irreversibly inhibited by C-difluoromethyl-3,4-dihydroxyphenylalanine (29) while histidine decarboxylase, ornithine decarboxylase and aromatic amino acid decarboxylase have been inhibited by the corresponding <=d-monof luoromethylanri.no acids, respectively (29). [Pg.248]

Recidivation is generally due to undetected small intra-hepatic foci. Especially in cirrhosis, the recurrence rate is very high as a result of this factor. A further cause of recidivation after primary RO resection is attributed to the multicentricity of the HCC, i. e. synchronic or metachronic development of additional tumours which are independent of the primary tumour. Therefore, the secondary prophylaxis takes on a special meaning (systemic or intra-arterial chemotherapy, interferon, retinoids, autologous lymphocyte transfusion, etc.). An increase in ornithine decarboxylase and spermidine is apparently a high risk factor for recurrence. Even after recidivation, repeated resection or interventional procedures (e.g. RFTA) are sometimes successful. [Pg.783]

Our microarray data set contains substantial information about the mechanism and extent of CS effects on various genes. The small number of distinct temporal patterns indicates that a limited number of mechanisms may mediate CS pharma-cogenomics. Of special note, several genes such as ornithine decarboxylase and hydroxysteroid sulfotransferase were represented by multiple different probes on the gene arrays. Reasonable concordance in prohles and dynamic parameters from multiple probes indicated a good degree of reproducibility in results. [Pg.522]

The use of suicide inhibitors of ornithine decarboxylase and arginine decarboxylase, coupled with the efficiency of incorporation of labelled precursors, led to the conclusion that arginine decarboxylase was the source of putrescine for tobacco alkaloids including 1 and 2 [46], Labelled spermidine was incorporated into 1 and 2, apparently via degradation to putrescine [47]. In vitro properties of the nicotine demethylase activity from Nicotiana otophora were characterized [59]. [Pg.183]

Bateman, 1994). Coleman and colleagues (1994) have documented rapid exchange between subunits of dimeric ornithine decarboxylase and suggest that this exchange facilitates regulation by antizyme. In contrast, there is no evidence for a rapid equilibrium between subunits of the 14-subunit chaperone GroEL (A. Horwich, personal communication), or in a number of other multisubunit enzymes (Distefano et al., 1990 Perry et al., 1992 Wente and Schachman, 1987). The potential importance of subunit exchange to sHsp function is discussed further below. [Pg.125]

There is some evidence to suggest that soluble compounds produced by lactic acid bacteria may act directly on tumor cells to inhibit their growth. Arimochi et al. (1997) showed (a) an inhibitory effect of L. acidophilus on azoxymethane-induced ACF formation in rat colon and (b) enhanced removal of 0 -methylguanine from the colonic mucosal DNA. This inhibitory effect was found in culture supernatants and not from bacterial cells. Furthermore, other studies have shown that dietary administration of lyophilized cultures of B. longum suppressed azoxymethane-induced colonic tumor development, along with a decrease in colonic mucosal cell proliferation and colonic mucosal and tumor ornithine decarboxylase and ras-p21 activities (Reddy, 1998). [Pg.758]
