For the kinetic resolution of racemic a-acetoxyamide 6 several native enzymes were used (Scheme 5.5). The native Upases from Pseudomonas cepacia (PCL) and porcine pancreas (PPL) showed the highesL although stiU unsatisfactory, enantios-electivity ( = 5.1 and 3.5, respectively). Upon immobilization into a solgel matrix, the enantioselectivity of PCL was improved significantly to 30.5. Most kidney transplant procedures are heterotopic, where the kidney is implanted above the pelvic bone and attached to the patient s iliac artery and vein. The ureter of the transplant kidney is attached directly to the recipient s bladder or native ureter. Banting, Sir Frederick. (1891-1941). A native of Ontario, Canada, Banting did his most important work in endocrinology. His brilliant research culminated in the preparation of the antidiabetic hormone that he called insulin, derived from the isles of Langerhans in the pancreas. He received the Nobel Prize in medicine for this work together with MacLeod of the University of Toronto. In 1930, the Banting Institute was founded in Toronto. Insulin is synthesized as preproinsulin that is proteolytically processed in the P cells of the islets of Lang-erhans in the pancreas, to give proinsulin. After synthesis and folding, a section of the molecule (the C peptide) is excised, leaving the A and B peptides connected via disulfide bonds. Rational protein engineering has been successfully used in a variety of problems of interest in biomedical engineering. One of the first, and perhaps the most widely used examples can be found with the protein engineering of human insulin for the treatment of diabetes [ 3 ]. Native insulin has evolved to form dimers and hexamers, so that it can be produced and stockpiled in the pancreas before it is needed for release in the body. When purified insulin is injected subcutaneously following a meal as a treatment for diabetes, only the active monomer form is desired, and thus the formation of dimers and hexamers can slow absorption. This has been addressed using site-directed mutagenesis to introduce repulsive charges and steric hindrances at the dimer interface, in order to reduce the tendency of human insulin to self-assemble. This work has led to insulin monomers that have an increased rate of absorption, and thus a produce a preferable postprandial plasma concentration profile [4]. [Pg.219]
