The prototype of a pure ethylenediamine is tripeleimamine antazoline [91-75-8] C27H22N2, belongs to the same family of compounds. Several well-known alkylamines in addition to chlorpheniramine are known. Hexchlorpheniramine maleate [2438-32-6] and triprohdine monohydrochloride monohydrate [6138-79-0] an alkenyl derivative, are two examples. 

Eor colorfastness to perspiration, ISO 10S-E04, the specimen is immersed in a solution of 0.5 g/L of 1-histidine monohydrochloride monohydrate and 5 g/L sodium chloride buffered to either pH 8.0 (alkaH perspiration test) or pH 5.5 (acid perspiration test) in a dish at 50 1 Hquor-to-goods ratio, at room temperature for 30 min. This is prepared by the reaction of N, N -dibenzylidenelysine methyl ester (124) with chlorodifluoromethane (HCCIF2) in the presence of lithium diisopropy-lamide. The resulting ester is hydrolysed with HCl to get DL-DFMO, which is isolated as its monohydrochloride, monohydrate [80]. 

To a solution of NaOMe (5.4 g, 0.1 mol) in 2-ethoxyethanol (200 mL) was added guanidine hydrochloride (9.6 g, 0.1 mol) and 2,4-diaminopyrimidine-5-carbonitrile (13.5 g, 0.1 mol). The mixture was then stirred at reflux for 20 h. During this time NH3 was continuously evolved. The hot mixture was filtered and the crude product washed with warm H20 (100 mL). Purification was effected by recrystallization (with charcoal) from H20 (600 nlL) containing coned HC1 (20 mL). An analytical sample of the free base was prepared by adding a small amount of the product obtained (the monohydrochloride monohydrate) to NH40H and then recrystallizing the resulting product from H/> yield 68% mp > 300°C.

A mixture of 1.8 parts of 1 -(2-chloroethyl )-4-ethyl-1,4-dihydro-5H-tetrazol-5-one, 3.45 parts of N-[4-(methoxymethyl)-4-piperidinyl] -N-phenylpropanamide, 5 parts of sodium carbonate, 0.2 part of potassium iodide and 240 partsof 4-methyl-2-pentanone is stirred and refluxed overnight with water-separator. The reaction mixture is poured onto water and the layers are separated. The organic phase is dried, filtered and evaporated. The residue is purified by column-chromatography over silica gel using a mixture of trichloromethane and methanol (97 3 by volume) as eluent. The pure fractions are collected and the eluent is evaporated. The residue is converted into the hydrochloride salt in 2-propanone. The salt is filtered off and crystallized from 2-propanone,yielding 15 parts (33.3%) of N-[1 -[2-(4-ethyl-4,5-dihydro-6-oxo-1 H-tetra-zol-1-yl] -4-(methoxymethyl)-4-piperidinyl] -N-phenylpropanamide monohydrochloride monohydrate melting point 140 °C.
