Molecular genetics research

The wide range of molecular techniques available and the rapid pace of new developments make it difficult for an individual to stay apprised of all aspects of the field that might impinge on the research at hand. For this reason, other researchers involved in molecular genetics are a key element of the local environment. This may include other members of the same research group, scientists who share the laboratory facility, or others doing various kinds of molecular genetics research at the same or nearby institutions. The collective molecular expertise in the local environment (and how well one can hope to make use of it) should be carefully assessed, and it may influence decisions on the choice of technique and the scope of research envisioned.

Bacteriophage X is a genetically complex and extensively studied virus of E. coli. Because it has been the object of molecular genetic research, it was investigated and developed as a vector. The DNA of phage X, in the form in which it is isolated from the phage particle, is a linear duplex molecule of about 45.5 kb pairs. The entire DNA sequence has been determined (SI). At each end are short, single-stranded 5 projections of 12 nucleotides that are complementary in sequence and by which the DNA adopts a circular structure when it is injected into the host cell i.e., X DNA naturally has cohesive termini that associate to form the cos site.

DNA gel electrophoresis is one of the most widely used tools of molecular genetics research. The biased reptation model has proven to be useful to understand most continuous field and low-frequency pulsed field effects. In this paper, we review how we have generalized the reptation model to include the electric forces acting on the DNA polymer, and we give an analysis of the main results obtained from analytical and numerical calculations. 

In this chapter we first discuss the development of systems for delivering DNA to plant cells. The remainder of the chapter then outlines the ways in which molecular genetics can be applied to research on stress tolerance, giving examples where these techniques are being used.
