Standards methodology

Cinnamic acid undergoes reactions that are typical of an aromatic carboxyhc acid. Using standard methodology, simple esters are easily prepared and salts are formed upon neutrali2ation with the appropriate base. Hydrogenation of cinnamic acid under mild conditions leads to 3-phenylpropanoic acid [501-52-0] whereas under forcing conditions, such as under high pressure in presence of a nickel catalyst, complete saturation to 3-cyclohexylpropanoic acid [701-97-3] is readily accompHshed (8). 

However, there is a standard methodology that ean be applied to the design of most eontrol systems. The steps in this methodology are shown in Figure 1.14. 

In other cases, attempts to cyclize the hydrazone proved more difficult and resulted in mixtures of compounds. The products obtained along with pyrazol-3-ones were ring-opened compounds or other pyrazole derivatives. Pilgram, who obtained hydrazones 56a,b by standard methodology, reported one such example.

The final goal of models used for testing or screening is to verify whether a special condition or feature is present or not. In this case the model system should be selected a priori based on scientific consensus and the testing protocols must be standardized in order to obtain comparable values when the same test is performed by two different laboratories or operators. Moreover, the existence of standardized methodologies could be very useful for regulators which need a certain degree of reliability to make appropriated decisions. 

The application of enzymes for the mild hydrolysis of esters has been well established as a convenient method, and hence can be regarded as a standard methodology in organic synthesis. Accordingly, the expectation is met that such enzymatic ester cleavage reactions are included in domino processes. Directed asymmetric reduction of a ketone has been brought about by the use of an intramolecular homochiral boronate ester250. The latter was readily introduced at a hydroxyl group in the molecule and has allowed the production of the enantiomeric alcohol, from the ketone by use of BH3-complex as the reductant (equation 64). Aspects such as the detector design, pretreatment, injection/separa-tion control and running buffer are the basis of a good performance of the microchips.
