Modem dmg development

MISRA R (1998) Modem dmg development from traditional medicinal plants using radiohgand receptor-hinding assays. Med Res Rev. 18 383-402. [Pg.181]

Although the use of bioactive natural products as herbal drug preparations dates back hundreds, even thousands, of years ago, their application as isolated and characterized compounds to modern drug discovery and development only started in the 19th cenmry, the dawn of the chemotherapy era. It has been well documented that natural products played critical roles in modem dmg development, especially for antibacterial and antitumor agents. More importantly, natural products presented scientists with unique chemical stmctures, which are beyond human imagination most of the time, and inspired scientists to pursue new chemical entities with completely different stmctures from known dmgs. [Pg.472]

Summarizing this chapter, it can be stated that the linking of NMR spectrometers (from 400 to 750 MHz) to various separation techniques has been developed into rather a powerful tool. Stereochemical problems in metabolism studies can be easily solved if the concentration of the present compounds is above the detection threshold. The coupling of capillary separation techniques together with NMR detection will stimulate high-throughput screening applications in modem dmg development. [Pg.116]

Although there are distinct difficulties with tritium, this isotope is an indispensable tool in modem dmg development. However, because of the risk of metabolic instability of tritium labels, tritiated targets require careful design and testing to ensure their suitability. [Pg.32]

Nature has been a potential source of therapeutic agents for thousands of years. An impressive number of modem dmgs have been derived from natural sources. Over the last century, a number of top selling dmgs have been developed from natural products. Anticancer dmg vincristine from Vinca rosea, narcotic analgesic morphine from Papaver somniferum, antimalarial dmg artemisinin from Artemisia annua, anticancer dmg Taxol from Taxus brevifolia and antibiotic peniciUins from Penicillium ssp. are just a few examples. [Pg.284]

The application of combinatorial chemistry to the synthesis of potential therapeutic agents has received increasing attention such that combinatorial chemistry is now an important tool in modem dmg discovery [71]. Automated approaches capable of screening large libraries of small molecules have resulted in the successful application of LC/MS in combinatorial chemistry. Current trends for further integration of LC/MS techniques with new instmmental development have generated structure-based assays for drug discovery. [Pg.315]

There is nonetheless little hope that the phase T-TTT aphorism will die. Nevertheless, it is quite wrong to assume that these classical terms and definitions still apply to how drugs are developed according to modem practice. The classical four-phase strategy of dmg development is far too stereotyped,... [Pg.102]

These modem aspects of dmg development are rarely optional. All are cmcial for the success of a product in the marketplace. Several of the next chapters will also describe methodologies that also teach us, on new dimensions, about properties that are intrinsic to dmgs. Some (e.g. pharmaceoeconomics) are also becoming of increasing importance in the regulatory approval process. [Pg.277]

Quite recently, attention has been drawn to the environmental contamination caused by pharmaceutical products and drugs. These groups constitute an enormous number of different substances, selected and/or manufactured for the purpose of biochemical and physiological activity and interference. By definition, such substances can act as toxicants in elevated concentrations and especially in the wrong situations or places. Furthermore, modem dmg design in many cases is directed towards the development of products with high bioavailability and... [Pg.85]

One important step in the development of any in vitro model is to correlate in vitro and in vivo data in order to validate experimental models and to assess the predictive power of the techniques (Pardiidge et al., 1990). Reproducibility and predictive ability of new models are of paramonnt importance, and uniform detailed validation guidance is necessary for the industry to accelerate the adoption of valid modem science in dmg development. [Pg.89]

As outlined above, the R D efforts of the modem pharmaceutical industry are affected by myriad factors including the political, social, and financial climates of the developed and developing world. Given that dmg development cycles are frequently of the order of ten years, it is difficult to predict which of these issues may have an impact on future performance and how to react to them. In addition, the pharmaceutical market is tmly international and so company management must monitor developments in numerous countries where they operate. However, it is clear that a few factors will be significant in the next decade. [Pg.38]

Nevertheless the gap between synthetic polymers and biomedicine, and especially the attempt to develop modem dmgs, is not yet closed. In contrast to this, the road to polymer technology and to the development of essential materials seems to be much easier. One of the reasons might be that in our universities we stiU teach chemistry... [Pg.17]

Lichens and other natural products remain as an untapped reservoir of potentially useful chemical compounds not only as drugs but also as unique templates that could serve as a starting point for synthetic analogues. Over 50 % of all modem clinical dmgs are of natural product origin, and natural products play an important role in the dmg development programmes in pharmaceutical industries. [Pg.21]
