Risk factors for the development of AML include exposure to environmental toxins, Hispanic ethnicity, and genetics.6 Of greater concern is the increased prevalence of AML as a secondary malignancy, resulting from chemotherapy and radiation treatment for other cancers. Alkylating agents, such as ifosfamide and cyclophosphamide, and topoisomerase inhibitors, such as etoposide, are linked to an increased risk of myelodysplastic syndrome (MDS) and AML.8... [Pg.1399]

The childhood tumors, such as Wilms s tumor and retinoblastoma, are a special group. They occur in infancy or childhood and apparently have a uniform incidence among all racial and geographic groups. Some cases are clearly inherited others are sporadic. One hypothesis is that the malignancy results from two mutations. If both mutations are somatic, that accounts for the sporadic cases. In retinoblastoma, mutations frequently affect only one eye. In the hereditary cases, one mutant gene is inherited from a parent and the second occurs somatically usually both eyes are affected. If this simple hypothesis is correct, the mutation component is the fraction of hereditary cases, estimated to be about one-third of all cases.21 ... [Pg.181]
