The lipid solubility, degree of ionization, pKa of the drug, pH of the drug solution, presence of saliva and the membrane characteristics, molecular weight and size of the drug, various physicochemical properties of the formulation, and the presence or absence of permeation enhancers, all affect the absorption and the permeation of drugs through the oral mucosa. [Pg.182]

Azone (laurocapram) is used extensively as a transdermal permeation enhancer, and has also found use in buccal drug delivery. It is a lipophilic surfactant in nature (Figure 10.4). Permeation of salicylic acid was enhanced by the pre-application of an Azone emulsion in vivo in a keratinized hamster cheek pouch model [35]. Octreotide and some hydrophobic compounds absorption have also been improved by the use of Azone [36], Azone was shown to interact with the lipid domains and alter the molecular moment on the surface of the bilayers [37], In skin it has been proposed that Azone was able to form ion pairs with anionic drugs to promote their permeation [38],... [Pg.208]

Some of these low molecular mass permeation enhancers act as surfactants/ detergents to increase the transcelluar transport of more lipophilic drugs by disrupting the structure of the lipid bilayer and rendering the cell membrane... [Pg.86]

Dermac SR-38 is one of a series of oxazolidinones, cyclic urethane compounds, evaluated as transdermal enhancers. The compound was designed to mimic natural skin lipids (such as ceramides), to be nonirritating, and to be rapidly cleared from the systemic circulation following absorption. In animal and human safety studies, Dermac SR-38 demonstrated a good skin tolerance (no observed irritancy or sensitization at levels of 1-10 wt% moderate to severe irritation in rabbit at 100%), and a low degree of acute toxicity (LD50(rat oral) > 5.0g/kg). The compound was evaluated for its ability to enhance the human skin permeation of diverse drugs from dermal and transdermal delivery systems. Data for minoxidil indicated an enhancer concentration-dependent effect for permeation enhancement. Dermac SR-38 was also found to enhance the skin retention of both retinoic acid when applied in Retin A cream, and dihydroxyacetone when applied in a hydrophilic cream. ... [Pg.1314]

Extent of Damage to Mucosal Cells. Permeation enhancement implies possible alteration of the protective permeability barrier either by 1) an increase in the fludity of intercellular lipids (relatively non-toxic) and/ or 2) extraction of intercellular lipids or denaturation of cellular proteins (much more damaging/toxic). Therefore, it is imperative that the permeation enhancer 1) exert a reversible effect 2) not be systemically absorbed and 3) not cause cumulative toxicity or permanent changes in the barrier properties. Application of up to 1% sodium lauryl sulfate or cetylpyridinium chloride to the ventral surface of the tongue of dogs resulted in desquamation, widening and separation of keratin.f The buccal mucosa of rabbits treated with... [Pg.2672]

Different mechanisms of absorption enhancement for these permeation enhancers have been proposed. Most permeation enhancers are thought to disrupt the lipid bilayer, which increases membrane fluidity. Some enhancers (non-ionic and presumably ionic... [Pg.2674]

Low molecular mass (LMM) permeation enhancers include various classes of substances. All molecules that interact with the membrane lipids or proteins and lead to membrane perturbation, in principle, be applied and may be helpful to guide drugs through the cells of the mucosa. However, the paracellular route... [Pg.1376]
