For lipid dynamics studies, the probe should be rigid, preferably rod- or disk-shaped with at least biaxial symmetry so that information on order can be obtained. [Pg.246]

The anthroylstearate series of fluorescent probes can be used to give information on lipid dynamics at different depths into the lipid bilayer.( 74) Compared with DPH, the anthroylstearates are less fluorescent and higher probe lipid ratios (1 100 compared to <1 400 with DPH) are needed. Although it is possible to extract time-resolved fluorescence anisotropy parameters with the anthroylstearates, so far studies have been largely confined to model lipid bilayers.(8 75 76)... [Pg.248]

Another series of probes which has attracted much interest in membrane studies has been the 2-A-(4-nitrobenzo-2-oxa-l,3-diazole) (NBD)-labeled lipids. These have been used for studies of the lipid trafficking in intact cells using mainly fluorescence microscopic techniques(77), for fluorescence quenching 78-791 and for studies of phospholipase A2 activities in membranes.(80) To date, they have received less attention in the study of lipid dynamics. [Pg.248]

The food supply is strongly influenced by the temperature of the water where the fish live. For this reason, the amplitude of metabolic variations during the annual cycle of a given fish is specific for each climatic zone, whether tropical, temperate-warm, temperate-boreal or polar. The seasonal variations in metabolism (e.g. lipid dynamics) display the greatest range in the... [Pg.106]

Such an approach was developed independently by the authors and their colleagues, who were prevented by international problems from becoming fully aware of one another s progress. With the introduction of openness in the former Soviet Union, it has been possible for the authors to meet and engage in full collaboration. This volume offers insight into hitherto inaccessible Russian data that concern principally the fatty fish and their lipid dynamics to this is added complementary western studies on non-fatty fish, especially their proteins. [Pg.360]

Piperine (1-piperoyl piperidine) in black pepper is shown to possess bioavailabilityenhancing activity with various structurally and therapeutically diverse drugs. This property of piperine may be attributed to increased absorption, which may be due to alteration in membrane lipid dynamics and a change in the conformation of enzymes in the intestine (Khajuria et al., 2002). [Pg.17]

Cell membranes are two-dimensional fluids that exhibit a wide range of dynamic behaviors. Recent technical advances have enabled unprecedented views of membrane dynamics in living cells. In this technical review, we provide a brief overview of three well-studied examples of membrane dynamics lateral diffusion of proteins and lipids in the plane of the membrane, vesicular trafficking between intracellular compartments, and exchange of proteins on and off membranes. We then discuss experimental approaches to monitor membrane protein and lipid dynamics, and we place a special emphasis on the use of genetically encoded fluorescent probes and live cell-imaging techniques. [Pg.197]

Generally speaking, two principal mechanisms operate in the biology of membrane processes, such as membrane transport and permeation. One involves a network of active sites and operates by metabolic energy, and it is referred to as active another is directed by passive diffusion, and it is called passive. This passive mechanism is determined by various aspects of lipid dynamics and lipid-protein interactions, and it can be described in quantitative terms of chemical and phase equilibrium and molecular physics. However, in the highly anisotropic... [Pg.1003]

In general, studies of fish lipid dynamics indicate that dramatic changes in total lipid are mainly accounted for by changes in non-polar (mainly TAGs) rather than polar lipids (e.g. phospholipids)12. The much smaller changes in tissue phospholipid levels may reflect the integral role of these lipids in membrane structure, and thus the necessity to conserve phospholipid concentrations within a relatively narrow... [Pg.126]

In sum, despite the little work that has been conducted on interspecies differences in total body lipid, lipid distribution and egg lipid, we feel it is a potentially important topic for predicting vulnerabilities to POPs in field-caught fish. Improved understanding of tissue lipid dynamics and lipid composition is likely to be important in any target species selected for field study of POP exposure and effects. [Pg.143]

The results summarized here establish that the additional motion that accompanies the -250 K dynamical transition of PM is required for proper BR function, and that the transition is hydration- and lipid-dependent. They also show that, in contrast to the case of soluble proteins, the PM transition is not coincident with the hydration water transition. Thus, the water and protein-lipid dynamics do not appear to be directly coupled as for soluble proteins. It should be clear from this summary that much remains to be done to develop a picture of membrane protein dynamics that is consonant with our current understanding of soluble protein dynamics. We now proceed to a comparison of protein-water dynamical coupling in the soluble protein MBP and the membrane protein BR, in the context of dynamical transitions. [Pg.377]
