Leukocyte subpopulations

Monocyte Monocytes are a class of white blood cells that co-purify with lymphocytes in commonly used density gradient procedures. They tend to be promiscuously sticky for the nonspecific (Fc) ends of monoclonal antibodies and therefore can lead to misleading results in analysis of leukocyte subpopulations unless their Fc receptors are blocked in the staining procedure. Monocytes differ from lymphocytes in their forward and side scatter characteristics.

Chemokines. Large family of small secreted proteins (8-15 kilodaltons) that control the trafficking of leukocyte subpopulations, induce leukocyte activation, and control - lymphocyte differentiation and effector function. May play an important role in the pathogenesis of autoimmune diseases, because the migration and accumulation of leukocytes in the target organs are critical steps for this.

Chemokine receptors are differentially expressed on leukocyte subpopulations dictating their ability to migrate both in physiological and pathological conditions. Their expression is modulated during leukocyte differentiation and maturation and they can be used as markers to identify and characterize the frequency and the activation state of...

In the last years, flow cytometry has become the technique of choice to detect chemokine receptors expressed by leukocytes. Added values of this technique include the possibihty to analyze chemokine receptors expression also on rare cell populations with no need of purification procedures when combinations of different antibodies identifying leukocyte subpopulations... 

The recently described chemokines ELC and SLC are specifically chemoattractive for lymphocytes and not other leukocyte subpopulations (55). Both are functional ligands for CCR7 (10,56), a receptor formerly known as EBIl (40,41). CCR7 is selectively expressed on activated T and B lymphocytes and is strongly upregulated upon Epstein-Barr virus infection of Burkitt s lymphoma cells (40).
