Oevere toxicological responses have been associated with certain chloro- dibenzodioxins. One of these responses is chloracne, a folliculosis first associated with skin contamination by chlorohydrocarbons in 1899 (3). Serious outbreaks of chloracne-like lesions associated with runaway reactions in the production of 2,4,5-trichlorophenol occurred in Germany in the early 1950 s (5). 2,4,5-Trichlorophenol itself does not cause acne (S), but the contaminants which may be formed in the uncontrolled production of 2,4,5-trichlorophenol are extremely potent acnegens (5). 2,3,7,8-Tetrachlorodibenzo-p-dioxin and tri- and tetra-... [Pg.55]

The most important survival and growth factor for mast cells is the KIT Ugand stem cell factor (SCF) [12], The hypothesis of early studies, that SCF might be elevated in skin lesions associated with mastocytosis [13], however, was not confirmed by later studies on SCF levels in skin and blood, at least for adult patients [14],... [Pg.111]

In 1998, the FDA approved the use of thalidomide for the treatment of lesions associated with erythema nodosum leprosum. Because of thalidomide s potential for causing birth defects, the distribution of thalidomide was permitted only under tightly controlled conditions. Nevertheless, because of its use for patients with leprosy thalidomide has been identified again as a current teratogen, now in South America. [Pg.419]

Gardasil was approved by the FDA in 2006. It is a quadrivalent recombinant vaccine against the human papilloma vims (HPV), more specifically against types 6, 11, 16 and 18. It is able to reduce pre-cancerous cervical, vaginal and vulvar lesions, associated with HPV types 16 and 18, as well as condylomas associated with HPV types 6 and 11. With the approval of the first HPV vaccine, cervical cancer now has a primary prevention tool. [Pg.462]

Production of PCBs increased between 1930 and 1970, with a large increase in production after World War II. It is estimated that 650,000 tons of PCBs were produced in the United States during this 40-year period, with a peak of 42,000 tons in 1970. Global production was approximately twice that of the United States during this same period. Another 400,000 tons have been produced outside the United States after their production was banned in this country in 1979. Health problems started to appear in PCB plant workers soon after their commercialization. The first problem observed among PCB workers was chloracne, a condition resulting in skin lesions associated with overexposure to chlorine compounds. [Pg.44]

Taking into account the essential role of PAL activity in the appearance of RS, Peiser et al. [146] studied whether the application of various inhibitors of PAL activity could prevent the development of RS lesions in lettuce midribs. The use of the inhibitor 2-aminoindan-2-phosphonic acid (AIP) sharply diminished the formation of phenolic compounds, although this did not reduce the number of lesions associated with RS [146]. In short, according to the results of Peiser et al. [146], the early development of RS lesions is independent of the increase in PAL activity and phenolic compounds, rather than resulting from these increases as previously suggested. However, the accumulation of phenolic compounds does contribute to the subsequent browning symptoms indicative of RS. [Pg.675]

Because of the rather nonspecific blockade of dopaminergic receptors, antipsychotics are associated with several adverse side effects. The most serious of these are abnormal movement patterns that resemble tardive dyskinesia, Parkinson disease, and other lesions associated with the extrapyramidal system. In some cases, these aberrant motor activi-... [Pg.102]

Besides the paranoid psychosis associated with chronic use of amphetamines, a specific lesion associated with chronic amphetamine use is necrotizing arteritis, which may involve many small and medium-sized arteries and lead to fatal brain hemorrhage or renal failure. Overdoses of amphetamines are rarely fatal they can usually be managed by sedating the patient with benzodiazepines. [Pg.731]

The retinoid-like compound bexarotene (63) is approved for treating skin lesions associated with T-cell lymphomas. The starting tetralin (60) is probably obtained by alkylation of toluene with dichloride (54). Fiiedel-Crafts acylation with the acid chloride (57), gives the ketone (61). This intermediate is then treated with the ylide from triphenylmethyl-phosphonium bromide. The carbonyl oxygen in the product (62) is now replaced by a methylene group. Saponfication of the ester affords the free acid and thus 63. °... [Pg.76]

Cholangiodestmctive cholestasis is caused by bile duct obstruction which may be intrahepatic or extrahepatic. Bile duct injury may lead to sloughing of epithelial cells into the lumen, cell edema, and inflammation, which may contribute to obstruction (Treinen-Moslen, 2001 Plumlee, 2004). Chronic lesions associated with cholangiodestmctive cholestasis typically include bile duct prohferation and periductular fibrosis. Vanishing bile duct syndrome, characterized by a loss of bile ducts, has been seen in chronic cholestatic disease in humans (Zimmerman, 1999 Treinen-Moslen, 2001) and has been produced experimentally in dogs (Uchida, 1989). [Pg.553]
