Bacterial desiccation and survival of other species in dried soils and mineral powders has been reported (Bitton et al., 1976, Labeda et al., 1976 Dupler Baker, 1984 Moll Vestal, 1992). In many instances the authors did not report soil mineralogical characteristics however, Bitton et al. (1976) showed greater survival of Klebsiella aerogenes under desiccation stress when in soils dominated by montmorillonite as compared to kaolinite. Citrate lyase catalyzes the cleavage of citrate to oxaloacetate and acetate in the presence of Mg2+ or Mn2+, but in the presence of EDTA it catalyzes its synthesis. The enzyme is a complex of three subunits. The y-subunit functions as an acyl carrier protein (ACP). The a-subunit is an acyl transferase involved in citryl-ACP formation and the release of acetate, and the /8-subunit catalyzes the cleavage of the citryl-ACP intermediate to oxaloacetate and acetyl-ACP. Urease (urea amidohydrolase) is an enzyme first identified over a hundred years ago in bacterial extracts [22], The presence of urease is a virulence factor for some pathogenic bacteria [23,24], It is now known to occur also in plants, fungi, and invertebrates (see [24,25] for reviews). Urease from jack bean was the first enzyme to be crystallized, in 1926. Almost 50 years later its metal content was reexamined and it was found to contain two atoms of nickel per subunit [26]. Finally in 1995 the crystal structure of the enzyme from the enteric bacterium Klebsiella aerogenes was determined [27], Amino-acid sequence comparisons predict that the structures of the plant and bacterial enzymes are similar, although with different subunit arrangements. [Pg.234]

Variable temperature magnetic circular dichroism (MCD) studies have revealed similar electronic and magnetic properties for the nickel centres in Jack bean and Klebsiella aerogenes urease.
