Microdialysis allows direct intracerebral administration of the drugs that normally do not cross the blood-brain barrier (BBB) or are metabolized in the circulation. It was demonstrated earlier (Ozaki et al., 1988 Westerink et al., 1990) that MPP+, a toxic metabolite of 1-methyl-4-phenyl-l, 2,3,6-tetrahydropyridine (MPTP), has a biphasic effect on TH activity when infused via the microdialysis probe for 20 minutes. Since, however, this drug does not penetrate the BBB, it was difficult to investigate its effects in vivo. Microdialysis overcomes this problem and has enabled the testing of a range of other modulators that previously could be studied only on in vitro preparations.

The mammalian suprachiasmatic nuclei contain an endogenous pacemaker that generates daily rhythms in behaviour and secretion of hormones. They stimulate evening ACTH secretion in the rat (Cascio etal. 1987). Kalsbeek etal. (1996) used microdialysis-mediated intracerebral administration of the vasopressin V,-receptor antagonist to study the mechanisms underlying the circadian control of basic corticosterone release in the rat. By timed administration of the vasopressin antagonist divided equally over the day/night cycle, they were able to uncover the existence of an additional stimulatory input from the suprachiasmatic nucleus to the hypothalamo-pituitary-adrenal axis. 

The older hypothesis in which aluminium was said to be involved in the progress of Alzheimer s disease has generated much controversy [37]. It nevertheless inspired the development of early models for Alzheimer s disease. Intracerebral administration of aluminium in rabbits brings about neurofibrillary degeneration in several brain regions and a decrease of choline acetyl transferase in the striatum, but not in the cerebral cortex and hippocampus where the decrease mostly occurs in Alzheimer s disease [38], no amyloid plaques are formed and few changes in other neurotransmitter systems and behaviour have been reported [39]. 

Systemic or intracerebral administration of pilocarpine hydrochloride in high doses induces seizures in rodents. These seizures are characterized by a sequential development of behavioral patterns and electrographic activity [69]. The pilocarpine seizure induces status epilepticus like in humans mimicking the pathogenesis and progression of mesial temporal lobe epilepsy [70]. It can be useful to exploit these model properties to design new therapeutic approaches for treating refractory epilepsies.
