Human simian virus 1 vaccine

Various bacterial vectors have been used to express a number of bacterial B. pertussis, S. pneumoniae, Y. pestis, and L. monocytogenes), viral (herpesvirus, influenza virus, human immunodeficiency virus, simian immunodeficiency virus, and hepatitis B virus), and parasitic (5. mansoni, and L. major) antigens. Significant improvements in attenuation of bacteria, and the stability, localization, and expression levels of heterologous antigens are required to market the bacterial vector-based vaccines for use in humans or animals. [Pg.3910]

A more general example from virus vaccine production is the rigorous examination of tissue cultures to exclude contamination with infectious agents from the source animal or, in the cases of human diploid cells or cells from continuous cell lines, to detect cells with abnormal characteristics. Monkey kidney cell cultures are tested for simian herpes B virus, simian virus 40, mycoplasma and tubercle bacilli. Cultures of human diploid cells and continuous line cells are subjected to detailed kary-ological examination (examination of chromosomes by microscopy) to ensure that the cells have not undergone any changes likely to impair the quality of a vaccine or lead to undesirable side-effects. [Pg.409]

Much of the preclinical data generated with regard to these vaccines entailed the use of one of two animal model systems simian immunodeficiency virus infection of macaque monkeys and HIV infection of chimpanzees. Most of the positive results observed in such systems have been in association with the chimp-HI V model. However, no such system can replace actual testing in humans. [Pg.409]
