Histological responses

Histologic response is defined as improving inflammation and fibrosis as noted by pretreatment and posttreatment biopsy scores. 

Ad.TNF gene therapy is presently in clinical trials in patients receiving radiotherapy. An open-label, phase I, dose-escalation study of tumor necrosis factor-alpha (TNFerade) gene transfer with radiation therapy for locally advanced, recurrent, or metastatic solid tumors is currently accruing patients and has several endpoints (82), including toxicity and tolerable dose. Pharmacokinetics will be evaluated and biological correlates will determine the histologic response to therapy. 

As part of a multicenter, randomized, double-blind, placebo-controlled trial of ribavirin in 59 patients with hepatitis C virus infection, liver biopsies were studied for iron deposition (19). Increased total iron deposition, preferentially in hepatocytes, occurred during a 9-month course of ribavirin. The deposition had no apparent effect on the biochemical or histological response to ribavirin therapy.

Comparative leaf histological responses — Lesion development in fronds of P. aquilinum and leaves of P. vulgaris and H. annuus was very similar. In these three species initial injury was characterized by collapse of one to six cells on the adaxial epidermis. Normal turgid cells which comprised a continuous epidermis became flaccid witL an initial increase in cellular stain intensity. This tissue collapse resulted in a depression on the leaf surface. Underlying mesophyll tissues showed little or no distortion at this stage.

PVDF sutures showed a similar histologic response to a PP suture (Urban et al., 1994). At the end of 6 months implantation in adult mongrel dogs, PVDF sutures were encapsulated by a thin layer of newly-formed connective tissue with the absence of inflammatory cells. The explanted and cleaned PVDF sutures revealed no visible surface damage or degradation.

In summary, all of the studies discussed show that CSM ([NaCa(P03)3] ) fiber is biologically degradable and at the concentrations studied in intratracheal instillation and inhalation studies produce a minimal histological response. The toxicity of the fibrous material appears to be directly related to the durability and dimensions of the fiber (Stanton et al., 1977,1981 Kuschner, 1987). Thus the present study suggests that CSM ([NaCa(P03)3] ) fiber should have a low chronic toxicity potential since the fiber appeared to be nondurable in biological systems. ...
