Ciprofloxacin has been tested in a randomized, place-bo-controlled study 70 patients with mildly to moderately active UC were randomized to receive ciprofloxacin 250 mg b.i.d. or placebo for 14 days. At the end of the study, 70.5% of patients in the ciprofloxacin group versus 72% in the placebo group showed an improvement [31], Nevertheless, in a more recent randomized, placebo-controlled trial, ciprofloxacin was administered for 6 months to patients with active UC poorly responding to conventional therapy with steroids and mesalazine. At the end of the study, the treatment failure rate was 21% in the ciprofloxacin-treated group and 44% in the placebo group (p < 0.002). Studies are often one to two weeks in duration and routinely include body weight determinations, clinical observations, and gross necropsy findings. Additional antemortem studies may be performed as appropriate, especially in large animals (e.g., observation of local reactogenicity, pharmacokinetic evaluations, hematological and/or clinical biochemistry measurements). In our laboratory, the viability of excised porcine buccal mucosa was assessed using histological evaluation and a 3-[4,5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) biochemical assay which has previously been used in assessing the viability of excised buccal mucosa and cornea [49, 50], Histological evaluation of tissue demonstrated that the buccal epithelium appeared viable up to 9 h postmortem, and this was supported by the MTT biochemical assay, which indicated that viability was maintained for up to 12 h [80], Therefore, we recommend that all permeation experiments using porcine buccal mucosa be performed within 9-12 h of animal death.

Reproductive toxicity tests are not required to support Phase I clinical studies in men. Detailed histological evaluations of the male reproductive organs should be performed in the repeated-dose toxicity studies. Male fertility studies in the rodent, however, would be expected to support Phase III studies. 

Previous studies of tumor blood vessel response to radiation have relied upon the clearance of Xe and the quantity of RBCs (Cr-59) within tumors following treatment with radiation (3,6,8). Currently available methods allow direct, longitudinal observation of tumor blood volume and blood flow (10,11). More recent models include the tumor vascular window (11), power Doppler sonography (10), and histologic evaluation of tumor blood vessels (12,13). These models permit characterization of the dose-dependent response in tumor blood vessels.

A comprehensive histology evaluation was performed in this study, but the liver is the only tissue specifically mentioned as having been examined. Due to the total number of tissues examined (21) and route of exposure, it is probable that the gastrointestinal tract was examined but not discussed because no histopathologic changes were observed. 

Historically, in vivo biocompatibility has been qualitatively assessed by histological evaluation of explanted devices.6 The presence or absence of specific inflammatory and fibrotic cell types and the degree and duration of the incidence of these cells at the implant site can be correlated to arelative degree of success or failure in terms of tissue compatibility. However, this common implant, sacrifice, explant, and evaluate single-time-point-per-animal approach is quite limited in assessment of time-depen-dent phenomena (e.g., direct assay of molecular and cellular mediators) associated with the progression of the inflammatory response and the development of the FBR.

The main parameters commonly used for monitoring the course of an experimental skin and soft tissue infection are the death or survival of the animal, the development and progression of the local skin lesion, and the removal of organs for determination of bacterial loads, immune response analysis, or histological evaluation. Survival curves can be constructed by monitoring the time of death of individual mice after bacterial inoculation (see Note 8).
