Small particle size resins provide higher resolution, as demonstrated in Fig. 4.41. Low molecular weight polystyrene standards are better separated on a GIOOOHxl column packed with 5 /u,m resin than a GlOOOHg column packed with 10 /Ltm resin when compared in the same analysis time. Therefore, smaller particle size resins generally attain a better required resolution in a shorter time. In this context, SuperH columns are best, and Hhr and Hxl columns are second best. Most analyses have been carried out on these three series of H type columns. However, the performance of columns packed with smaller particle size resins is susceptible to some experimental conditions such as the sample concentration of solution, injection volume, and detector cell volume. They must be kept as low as possible to obtain the maximum resolution. Chain scissions of polymer molecules are also easier to occur in columns packed with smaller particle size resins. The flow rate should be kept low in order to prevent this problem, particularly in the analyses of high molecular weight polymers. [Pg.143]

The injection volume should be kept as small as possible to attain maximum resolution in analyses. This is particularly important in analyses on columns packed with small particle size resins such as SuperH. The H-type cell devised by Lingane and Laitinen and shown in Fig. 16.9 will be found satisfactory for many purposes a particular feature is the built-in reference electrode. Usually a saturated calomel electrode is employed, but if the presence of chloride ion is harmful a mercury(I) sulphate electrode (Hg/Hg2 S04 in potassium sulphate solution potential ca + 0.40 volts vs S.C.E.) may be used. It is usually designed to contain 10-50 mL of the sample solution in the left-hand compartment, but it can be constructed to accommodate a smaller volume down to 1 -2 mL. To avoid polarisation of the reference electrode the latter should be made of tubing at least 20 mm in diameter, but the dimensions of the solution compartment can be varied over wide limits. The compartments are separated by a cross-member filled with a 4 per cent agar-saturated potassium chloride gel, which is held in position by a medium-porosity sintered Pyrex glass disc (diameter at least 10 mm) placed as near the solution compartment as possible in order to facilitate de-aeration of the test solution. Independent studies involving two separate collections of A. cavernosa from Fiji afforded kalihinol-A (107), -F (112), -X (109), and a new compound, isokalihinol F (118) [65]. The latter was obtained from a batch, in which kalihinol-A was the major component. Recollection of the same sponge a year later, however, resulted in -X (109), -Y (110), and -Z (111) as the only kalihinols [65], It is unclear, whether seasonal variation is the determining factor of the chemical changes noted in A. cavernosa.
