The somewhat controversial theory of risk homeostasis is relevant to a discussion of risk taking. RHT was developed initially in the area of driving behavior (Wilde, 1984). The theory states that accident rates are not determined by actual levels of intrinsic risk but by the levels of risk acceptable to individuals in the situation. The theory implies that people adjust their risk-taking behavior to maintain a constant level of perceived risk. Thus, if improved safety measures are introduced (e.g., better guarding, improved protection systems then individuals will behave in a more risky fashion in order to maintain their accustomed levels of risk. [Pg.138]

Fogli et al. developed and validated an HPLC method with fluorescence detection for simultaneous routine TDM of anthracyclines and their metabolites.27 They coupled a Waters LC Module I Plus system equipped with a WISP 416 autosampler with a Model 474 scanning fluorescence spectrophotometer. The mobile phase consisted of 50mM monobasic sodium phosphate buffer and acetonitrile (65 35 v/v), adjusted to pH 4.0 with phosphoric acid. The flow rate was 1 mL/min. A 1-1., four-necked, round-bottomed flask equipped with reflux condenser, sealed stirrer, thermometer, and solid addition funnel and protected from atmospheric moisture with a Drierite guard tube is carefully dried and flushed with a dry inert gas (Note I). The flask is charged with 453 g. (3.1 moles) of silver difluoride (Note 2) and 500 ml. of l,l,2-trichloro-l,2,2-trifluoroethane (Note 3), and phenyl disulfide (100 g., 0.458 mole) (Note 4) is weighed into the solid addition funnel. The stirrer is started, and phenyl disulfide is added to the slurry in small portions. An exothermic reaction occurs, and after the addition of several portions the reaction mixture reaches a temperature of 40° (Note 5). By intermittent use of a cooling bath and by adjusting the rate of addition of the disulfide, the reaction temperature may be maintained between 35° and 40°. The addition of the phenyl disulfide requires 45-60 minutes. On completion of the addition the suspension of black silver difluoride has been converted to yellow silver monofluoride, and the exothermic reaction gradually subsides. The reaction mixture is stirred for an additional 15-30 minutes without external cooling and then quickly heated to reflux. [Pg.42]

Rezk et al. [74] developed and validated a reversed-phase HPLC assay method for the simultaneous quantitative determination of omeprazole and its three metabolites in human plasma. The method provides excellent chromatographic resolution and peak shape for the four components and the internal standard within a 17-min run time. The simple extraction method results in a clean baseline and relatively high extraction efficiency. The method was validated over the range of 2-2000 ng/ml. The resolution and analysis for the four analytes omeprazole, hydroxyome-prazole, omeprazole sulfone, and omeprazole sulfide and the internal standard utilized a Zorbax C18 (15 cm x 3 mm, 5 /im) with a Zorbax C18 (12.5 cm x 4.6 mm) guard column. The mobile phase consisted of two components. Mobile phase A was 22 mM phosphate monobasic, adjusted to a pH of 6 with diluted sodium hydroxide. This solution was filtered through a 0.45-/im membrane filter, then mixed as 900 ml buffer to 100 ml methanol. Mobile phase B was composed of 100 ml of the phosphate buffer as mobile phase A, mixed with 800 ml of acetonitrile, 100 ml of methanol, and 100 /A of trifluoroacetic acid with an initial flow-rate of 0.55 ml/min and detection at 302 nm. [Pg.219]
