Galactose-binding lectins

Limited data are available with respect to the carbohydrate-binding specificity of abrin. Olsnes and coworkers147 demonstrated that the agglutination of erythrocytes by abrin in the presence of anti-abrin antiserum was inhibited best by D-galactose. Lactose, melibiose, and D-fucose (in the order of decreasing potency) gave weaker inhibition, whereas D-glucose, D-mannose, D- and L-arabinose, D-xylose, 

D-fructose, D-ribose, and maltose were without effect. Equilibrium-dialysis analysis147 revealed one binding site for lactose on the abrin molecule, with an apparent association constant of 8 X 103 M l. 

A pH-dependent dissociation of the tetramer to dimers of molecular weight 64,000 occurred between pH 2 and 7. Analysis of the C-terminal amino acid showed both alanine and leucine, and the N-terminus was valine. The possibility of multiple isolectins was indicated by multiple band-formation of isoelectric-focused hemagglutinin.611

The disaccharide derivatives 196 and 197 of N-tosyl-L-serine, of interest for the study of D-galactose-binding lectins, were prepared by Kaifu and Osawa148 from 194 and 195, respectively, by protection of 0-4 and -6 with a benzylidene group, condensation of the acetal with tetra-O-acetyl-a-D-galactopyranosyl bromide (110) under Koenigs-Knorr conditions, and deprotection.
