Factor XIII. Factor XIII circulates in the blood as a zymogen composed of two pairs of different polypeptide chains designated A and B. Inert Factor XIII has a molecular weight of 350,000 daltons and is converted to its active transglutaminase form in the presence of thrombin and calcium. Activated Factor XIII, Xllla, induces an irreversible amide exchange reaction between the y-glutamine and S-lysine side chains of adjacent fibrin... [Pg.174]

Fibrin is formed from fibrinogen synthesized by the liver and secreted into the circulation. The conversion of fibrinogen to fibrin is initiated by a serine protease, thrombin. Thrombin, at the same time, can activate a transglutaminase enzyme, factor XIII present in... [Pg.503]

Fibrinolytics. Figure 2 Various fibrin structures for plasmin. Fibrinogen (Fg) is converted to fibrin (F) by thrombin (T), and thrombin can also convert factor XIII (XIII) to activated factor XIII (Xllla). The latter produces crosslinks between fibrins (FxxF) and also may crosslink fibrin with a2-plasmin inhibitor (FxxFxxPI). The efficiency of digestion of these plasmin substrates by plasmin, resulting in the soluble fibrin degradation products (FDP), is different. The amount of FDP formed in time is expressed in arbitrary units. [Pg.504]

Cryoprecipitate Component of blood obtained by freezing and thawing plasma that is rich in clotting factors. One unit of cryoprecipitate derived from a unit of whole blood contains a volume of 10 to 20 mL, 80 to 100 units of factor VIII which consists of both the procoagulant activity and the von Willebrand factor, 150 to 250 mg of fibrinogen, 50 to 100 units of factor XIII, and 50 to 60 mg of fibronectin. [Pg.1564]

A (very rare) genetic deficiency in the production of factor XIII also results in impaired clotting efficacy in affected persons. In this case, covalent links that normally characterize transformation of a soft clot into a hard clot are not formed. Factor XIII preparations, partially purified from human blood, are used to treat individuals with this condition to date, no recombinant version of the product has been commercialized. [Pg.340]

Factor XIII is present in both plasma and platelets. Plasma factor XIII is a 320 kDa tetramer, composed of two (70 kDa) a-chains, and two (90 kDa) /i-chains. The platelet form of factor XIII... [Pg.363]

In the case of platelet-derived factor XIII, the resultant product (a )2, is the activated form. Thrombin action on plasma-derived factor XIII generates an a 2jfi2 dimer, which is devoid of transglutaminase activity. However, in the presence of Ca +, the a P chains dissociate, yielding the biologically active a 2. [Pg.365]
