Enumeration media

The situation is somewhat less complicated when pertinent numbers of survivors are of the order 10 to 10 g because in this instance direct plating procedures can be used. With this technique the recovery of stressed cell populations presents fewer problems. Primary plating onto a non-selective solid growth medium, followed by replicating onto the selective enumeration medium, presents a solution in most cases .

Whatever medium is used as part of the agar, some bacteria that are present in the respective environment will not grow as they will be nonculturable. Cells in trypan blue viability stain were enumerated microscopically. The spleen cells were mixed with an equal number of SP/2.0 myeloma cells in the semi-log growth phase in RPMI medium. The cell mixture was centrifuged (200 x g) for 10 minutes and the cell pellet was suspended in 1 mL of polyethylene glycol (3 000 - 4 000 mol. wt. range) at 37 °C. The suspension was mixed continuously for one minute, followed by the addition of 1 mL of RPMI medium and another one minute of continuous mixing. An additional 9 mL of RPMI medium... 

Moreover, the structure of gels produced depends substantially on precipitation conditions and is determined by the precipitation temperature, pH of the medium, initial composition of solution, its concentration, of sequential mixing of solutions or conditions of introduction of precipitating agent [7]. It should be noted here that even in the production of one-component gel, periodicity of precipitation gives a product with a non-homogeneous structure which, as a rule, forms adsorbents with a set of pores different in size. Therefore, strict observance of all the conditions of sample synthesis enumerated above is absolutely necessary. 

Now we shall draw the reader s attention to an interesting fact. As shown in Sect-tion 2.3, the difference between the delocalized and solvated electron levels for hexamethylphosphotriamide is by almost the same value, i.e. about 0.4 eV higher than for water or liquid ammonia, i.e. for the solvents having a branched structure of H bonds. It follows that the introduction of a hydrocarbon residue into the solvent s molecule forces out only the delocalized electrons from the polar medium the solvated electron energy level in all the enumerated solvents has almost the same value. In samples with hydrophobic compounds, enumeration of SRB and general anaerobic bacteria (GAnB) may be compromised because of the low solubilization of these compounds in the culture medium. The study of the influence of surfactants in the SRB and GAnB estimation in oil samples may become an alternative to improve the MPN method under these conditions. Surfactants are organic compounds that present both a hydrophobic part and a hydrophilic part in the same molecule. This hydrophilic part allows surlaclants to be soluble in water, whereas the hydrophobic part causes them to concentrate at interfaces [4]. 

For GAnB enumeration, a medium was prepared under anaerobic conditions with the following composition (per liter) glucose, 5.0 g peptone, 4.0 g yeast extract, 1.0 g resazurin 4 ml (0.025% m/v), at pH 7.6. After preparation, the medium was distributed into 9 ml flasks and sterilized in an autoclave at 121 °C for 20 min. Then, sodium thioglycolate (12.4 g/I) was added, as previously described. 

Various organic solvents were used as reactionary medium at nonequilibrium polycondensation in solution realization [96]. The solvent type influence on the synthesis reaction main characteristics (conversion degree Q and molecular weight MM) is well known and is explained usually by solvent various characteristics (dielectric constant, solubility parameter, heat of dissolution and so on) [96]. However, up to now the indicated effects general theoretical explanation is not obtained. Besides, at the solvent type influence analysis its correlation with polycondensation process quantitative characteristics (the same Q and MM) is usually considered, but any changes of polymer structure or reaction mechanism are not assumed, although the possibility of side reactions is noted repeatedly [96]. The authors [71, 127] studied the solvent influence on the enumerated above characteristics on die example of the rules of chloranhydride of terephthalic acid and phenolfthaleine low-temperature polycondensation (polyarylate F-2), performed in 8 different solvents [128]. 

Hence, the stated above results have shown that the molecular weight, macromolecular coil stmcture, characterized by its fractal dimension, and medium, in which flocculation occurs, are included in a factors number, influencing on adsorption of admixtures (flocculation) effectiveness by a pol mier flocculator PDMDAAC. The medium cans influence on flocculation process by two ways by a macromolecular coil fractal dimension change and viscosity variation.
