Figure 21.4. Immuno-staining for cartilage specific collagen lype 11 molecules within matrix synthesized by embedded cultured human articular chondrocyte cells (500,000 cells per capsule). Staining of new matrix is strong throughout each sampled capsule. Individual cells are located in well-defined lacuane at a high density. New matrix is not formed in capsules containing primary hBMSC. Figure 21.4. It was made clear that his organization had a serious and deeply embedded culture problem, that no solution could be effective without making significant changes in operating procedure, and that the culture change needed could not be achieved without his direction and visible involvement over quite a long time. [Pg.325]

Industry has been looking for quick fix solutions for the last 10-15 years. Learning is not embedded in the culture—this is a business based on scientific endeavor with a core culture of individualism and heroic individuals. ... [Pg.259]

Fig 5 Immunogold labelling on thin sections of low-temperature embedded cell walls from 9-day old tobacco cells with JIM 5, a monoclonal antibody that recognises a relatively unesterified pectic epitope. Cell walls of elongating cells label very weakly, but material that is being secreted into the culture medium labels strongly. The old part of the wall is labelled but new wall material is not. [Pg.102]

Dorothy Nelkin Is there a gene for humour There s a gene for happiness - there must be one for humour. You know the only way I can figure out how to counter the impact of reductionism is to make people aware of the absurdity of what is happening, because certain assumptions become so embedded in culture and the way we think about things that they are not raised to the level of consciousness. Once people are aware then there s hope of thinking more seriously about the policy consequences. [Pg.318]

If the desire in examining cells in culture is to relate the findings to solid tissues, then the investigator may opt to test the cultured cells as though they were a solid tissue by embedding and sectioning them. [Pg.73]

The preparations most often affixed to silane- (9) or poly-L-lysine-coated slides are Carnoy-fixed suspensions of metaphase cells for chromosome studies (see Chapter 47) cytospins of cultured cells followed by methanol fixation sections of formalin-fixed, paraffin-embedded tissues and frozen sections fixed with acetone or ethanol/acetic acid after cutting. Each of these approaches imparts different intracellular effects that may modify cytological detail and/or hybridization. (See Chapters 8 and 9 for a discussion of fixatives and frozen-section preparations.)... [Pg.358]

Tissue culture cells are easy to handle under laboratory conditions. It is necessary to fix these cells immediately after cytospin or before paraffin embedding. Different fixatives are used depending on the goal of the experiment. [Pg.383]

For positive controls, it is extremely important to have a tissue culture cell line with an abundant amount of target nucleic acid. Varieties of tissue culture cell lines are available from American Tissue Culture Center, Rockville, MD. Rapidly growing tissue culture cell lines transfected with the target nucleic acid could be successfully used for positive controls. For example, formalin-fixed paraffin-embedded foreskin fibroblast tissue culture cell-line FS4 can be used as a positive control for human lysyl oxidase mRNA detection and c-W-ras transformed RS-485 cell line for normal ras message detection. Thin sections (4-5 im) of paraffin-embedded positive control cell lines could be placed simultaneously near the side of the experimental human or animal tissue. Similarly, one can also use freshly cut animal or human tissue (with the target sequences) preserved, sectioned, and mounted under standard laboratory conditions. For negative controls, choose a cell line (or a tissue) that completely lacks the target nucleic acid see Note 12). [Pg.384]

Cells of concavum adhered tenaciously to the surface of the culture flask and were embedded in mucilage. Only the most vigorous aspiration dispersed these aggregations sufficiently to permit reproducible counts. The mean dorsoventral diameter of cultured cells was 45 im. No morphological differences between axenic and bacterized cells were noted, but Irregularly-shaped cells were common in ESAW control cultures. [Pg.281]

Loeblich et al. (17) reported that rhathymum (=. mexl canum) was largely non-motlie and embedded in copious mucilage in their cultures and In the field. We have not found this either In our... [Pg.285]

Rotating culture vessels such as simulated microgravity systems are primarily used to study 3-D tumor growth and differentiation. However, mixed cell populations combined with matrix proteins can be used to generate a complex microenvironment in which cell-cell interactions and invasion can be measured (95). A similar system has also been described for the coculture of endothelial cells, myofibroblasts, and tumor cell clusters embedded in Matrigel . Differential labeling of the cell populations enables their invasion and the effects of inhibitors to be measured (96). [Pg.241]
