Electrophoresis of nucleic acids

Several research groups used another interesting column technology as an alternative to the modification of the capillary surface. This method is inherited from the field of electrophoresis of nucleic acids and involves capillaries filled with solutions of linear polymers. In contrast to the monolithic columns that will be discussed later in this review, the preparation of these pseudostationary phases need not be performed within the confines of the capillary. These materials, typically specifically designed copolymers [85-88] and modified den-drimers [89], exist as physically entangled polymer chains that effectively resemble highly swollen, chemically crosslinked gels. [Pg.25]

To understand how these modem methods work, it is necessary first to review some general laboratory techniques ubiquitous in genetic engineering. Among the most important are gel electrophoresis of nucleic acids, nucleic acid hybridization assays, and the polymerase chain reaction. [Pg.32]

Details of DNA electrophoresis methods are given in Table 9.3. The most widely used buffers in gel electrophoresis of nucleic acids are Tris/acetate/EDTA (TAE) and Tris/borate/EDTA (TBE). TBE has the best buffering capacity but the use of TAE tends to result in somewhat sharper bands. For some purposes, such as DNA extraction with glassmilk, TBE should be avoided unless sorbitol is... [Pg.188]

This chapter will describe the efforts taken to attempt to bring order to various electrophoretic methods by means of physical standards or standards activities. The discussion will not extend to electrophoresis of nucleic acids, which because of the nature of relative comparisons, needs to be addressed independently. [Pg.102]
