The use of HMDS as a derivatization reagent in the analysis of triterpenoid resins has been less explored. The TMS derivatives of triterpenoids bearing hydroxyl groups [a-amyrine, p-amyrine and hop-22(29)-en-3p-ol] have been identified in the triterpenic fraction of Burseraceae resins, thus demonstrating that HMDS combined with Py-GC/MS is effective in the derivatization of triterpenoid compounds [59]. However, the range of structures that can be fully derivatized and detected must be extended and, in order to get comprehensive results comparable with those coming from the well assessed off-line GC/MS procedures, general improvements in the on-line trimethylsilylation-pyrolysis method are needed. [Pg.342]

Moved] Cranberry fruit of Early Black cultivar was fractionated chromatographically and fractions were analyzed for flavonoid content. The effects of the flavonoid fractions and ursolic acid, an abundant triterpenoid in cranberry peel, were assessed in two models of colon cancer and one model of breast cancer. Clonogenic soft agar assays were used to determine the effect of these compounds on tumor colony formation in HCT-116, HT-29 and MCF-7 cells. MTT and trypan blue assays were performed to assess their ability to inhibit tumor cell proliferation. TUNEL assays were performed to assess apop-totic response to the cranberry compounds. The proanthocyanidins inhibited tumor colony formation in HCT-116 and HT-29 cells in a dose-dependent manner, with greater effect on the HCT-116 cell line. Ursolic acid strongly inhibited tumor colony formation in both colon cell lines. In addition to H and NMR analysis, TLC coupled in an offline manner with electrospray mass spectrometry (ESI-MS) and high-resolution ESI Fourier transform ion cyclotron resonance (FTICR) MS for isolation and characterization of two novel lupane triterpenoids from Paullinia pinnata L. 6P-(3 -methoxy-4 -hydroxybenzoyl)-lup-20(29)-ene-one and 6P-(3 -methoxy-4 -hydroxybenzoyl)-lup-20(29)-ene-ol, which are suspected to play a crucial role in the plant s wound healing effects [32]. The extract from Paullinia pinnata L. was subjected to an exhaustive isolation procedure starting with two consecutive column chromatography purifications on silica gel different ratios of chloroform-methanol were used as eluent. The triterpenoid-containing fraction was purified on preparative silica gel TLC plate with chloroform-methanol (9 1, v/v) as a developing solvent.
