Dissolved organic carbon analysis

Size exclusion chromatography (SEC) has been used to measure molecular weight (MW) distribution of humic substances (3, 6-9). Coupled with detection methods such as molecular fluorescence spectroscopy and dissolved organic carbon analysis (7), electrochemical detection (9), and atomic emission spectroscopy (5), SEC has been used extensively to study humic-metal complexes. A major disadvantage of SEC is that it does not provide adequate resolution for separating humic materials as they do not appear to be made up of distinct fractions with large differences in MW.

For aerobic degradation, uptake of oxygen or the evolution of carbon dioxide is most widely used. Use of the concentration of dissolved organic carbon may present technical problems when particulate matter is present, though analysis of dissolved inorganic carbon in a closed system has been advocated (Birch and Fletcher 1991), and may simultaneously overcome problems with poorly soluble or volatile compounds.

In both TOC and dissolved organic carbon (DOC) determinations, OC in the water sample is oxidized to produce carbon dioxide (C02), which is then measured by a detection system. Inorganic carbon (IC) is removed prior to the analysis by acidifying the sample. Alternatively, TOC can be determined indirectly through the measurement of total carbon (TC) and IC. TOC in the indirect method is calculated as the difference between the two. 

Reference conditions These are physical or chemical parameters that need to be assessed in conjunction with the analysis for standard compliance (e.g., temperature, pH, organic carbon content for soil or dissolved organic carbon, and total suspended solids for surface water) (depending on which factors influence chemical behavior and toxicity).
