The specific polysaccharide of the dominant O somatic antigen of Shigella dysenteriae has been shown to be electrophoretically and ultra-centrifugally homogeneous, and to possess a molecular weight of 26,000 from sedimentation and diffusion measurements.2601... 

One of the most serious obstacles in the phase equilibrium studies of polymer blends is the viscosity of the system. At temperahires limited by the degradation the self-diffusion coefficient of macromolecules is of the order of magnitude 10 to 10 m /s [Kausch and Tirrell, 1989]. As a result the phase separation is slow. To accelerate the process a low speed centrifuge, the centrifugal homogenizer (CH), with PICS has been used [Koningsveld et al, 1982]. In short, centrifugation within the immiscibility zone permits determination of binodal and critical points, while the use of PICS mode allows location of the spinodal.

Use of weak cation-exchange columns Supernatants of centrifuged homogenates can be passed through weak cation-exchange columns for prior separation of the basic catechol- and indoleamines (Warsh et al, 1979). 

Phase Volume Ratio Low Speed Centrifuge and Centrifugal Homogenizer. Measurement of the phase volume ratio (r, see p. 299) offers a simple but effective means of determining critical points. At a cloud point, r is infinite and zero, respectively, for concentrations above and below the critical concentration. At the critical concentration r = 1 at the cloud point (critical) temperature. Friday et al. also Wolf and Blaum have studied phase volume ratios for oligomer mixtures, in the latter case at elevated pressures.

Design Scope. Stirred vessels, rotor-stator mixers, static mixers, decanters, settlers, centrifuges, homogenizers, extraction colunms, and electrostatic coalescers are examples of industrial process equipment used to contact liquid-liquid systems. Although this chapter emphasizes stirred vessels, the fundamentals of phase behavior are applicable to a broad range of other equipment types. Immiscible liquid-liquid systems are processed in batch, continuous, and semicontinuous modes.
