Although these findings may apply to an experimental cortisone ulcer in animals, Hitzelberg in our laboratory obtained less definite results in man (H22, H23). Berg et al. (B5a) correlated the results of gastric biopsies with chemical and electrophoretic analysis of gastric juice. They determined total proteins by the biuret method and total hexoses colorimetrically by the orcinol technique, and performed horizontal paper electrophoresis followed by amido black and PAS staining. Their work corroborated previous findings (G19) that carbohydrate materials staining with PAS were localized in the center of the electrophoretic partition of gastric juice with some extension to adjacent areas. Arising from the decomposition of C -labeled carbohydrates are an enormous variety of products. For example, when a sample of D-glucose-C (about 6 mg., having a specific activity of about 14.44 mC. per millimole) was stored as a freeze-dried sample in the dark for 26 months, a 14.5% decomposition of the D-glucose occurred and, by use of two-dimensional paper chromatography-paper electrophoresis, the presence of 37 constituents was revealed. The greater complexity of this system in comparison with that of 7-irradiated solutions of D-glucose supports the view that direct-action effects supplement the decomposition caused by secondary-radiation effects, which are entirely responsible for the decomposition when dilute solutions are irradiated. 

Since 1958 when paper electrophoresis of serum proteins was reviewed in this series (P3), there have been no major technical developments in the art. The incorporation of nonionic detergents (Tween 80, Span 20, etc.) in the conventional barbiturate buffer (D14, G18, L14) and the use of tris -borate as buffer (A14, S27) have led to the separation of 7 to 11 globulin fractions. After zone electrophoresis in borate-containing buffer, an increased number of bands can be visualized by staining, but these are not revealed by the schlieren scanning technique after free electrophoresis in similar buffers (S27) the effects of this buffer may be due to interaction between borate and proteins having a high carbohydrate content.

Paper electrophoresis is, even in this age of high-performance liquid chromatography (h.p.l.c.), a useful method of analysis. The equipment required is inexpensive, and the technique is simple. It lends itself well to comparison of the coordinating ability of many carbohydrates with any one cation, or of that of several cations with any one carbohydrate.
