Glycosydation AChE and BChE carry 3 and 9, respectively, N-glycosylation consensus sequences attaching carbohydrate residues to the core protein via asparagines. Different molecular forms of the enzymes in various tissues, show different number and composition of carbohydrate residues. N-glycosylation at all sites was shown to be important for effective biosynthesis, secretion and clearance of ChEs from the circulation. Altered patterns of AChE glycosylation have been observed in the brain and cerebrospinal fluid of Alzheimer s disease (AD) patients, with potential diagnostic value.

The nature of the diet sets the basic pattern of metabohsm. There is a need to process the products of digestion of dietary carbohydrate, lipid, and protein. These are mainly glucose, fatty acids and glycerol, and amino acids, respectively. In ruminants (and to a lesser extent in other herbivores), dietary cellulose is fermented by symbiotic microorganisms to short-chain fatty acids (acetic, propionic, butyric), and metabohsm in these animals is adapted to use these fatty acids as major substrates. All the products of digestion are metabohzed to a common product, acetyl-CoA, which is then oxidized by the citric acid cycle (Figure 15-1). 

The basic form of ketosis occurs in starvation and involves depletion of available carbohydrate coupled with mobihzation of free fatty acids. This general pattern of metabohsm is exaggerated to produce the pathologic states found in diabetes meUitus, twin lamb disease, and ketosis in lactating catde. HPAEC analyses were carried out to determine the oligomeric products released from various pectic substrates after depolymerization by the PL isoenzymes. Action pattern analyses for the concerted action of PL isoenzymes utilized 68% esterified pectin as substrate. One-ml reaction mixtures in a buffer system as detailed in section 2.2. comprising 0.5% (w/v) substrate and 5 U of enzyme were incubated for 30 s to 18 h, and then thermoinactivated. Samples of 750 pi were applied to a Carbopac PA-1 (Dionex) column before the carbohydrates were eluted over a period of 70 min using a gradient of 0.2 M KOH, 0.05 M K-acetate to 0.2 M KOH, 0.7 M K-acetate. Detection employed a Pulsed Electrochemical Detector (PED, Dionex) in the integrated amperometry mode according to the manufacturer s recommendations. 

The presence of unusual carbohydrate fermentation patterns (particularly for lactose), and the ability to use citrate among Enterobacteriaceae has hindered, and sometimes jeopardized, the identification of pathogenic strains including Salmonella typhi.
