MDC biological activity is assayed by calcium flux or chemotaxis using either primary cells or more commonly stable transfectants expressing CCR4 (Imai et al, 1998). 

CCR3 Receptor Structure. Chimera studies of CCR3 and CCRl receptors show that the N-terminal segments of CCR3 appear to be important for eotaxin binding (77). However, eotaxin remained an effective agonist at this chimeric receptor in either calcium flux or chemotaxis assays. These data are consistent with a multisite model for chemokine-chemo-kine receptor interaction.

A second phase of the work made use of a radioligand assay for N-type channel binding in the chick brain. Using radiolabelled (o-conotoxin GVIA ((d-CTX), a peptide calcium channel antagonis, we confirmed the diversity of (o-agatoxin in funnel web spider venom and described a new class of toxins (Type III) which have broad specificity for vertebrate high threshold calcium channels. Subsequently, we used potassium stimulated synaptosomal calcium flux to discover toxins that block P-type calcium channels in the mammalian bndn.
