We have shown that a number of methods have been used in attempts to cal-eulate or prediet ligand affinity. Historieally, the methods that have been the most sueeessful are those which use the most target specific data, such as QSAR and LIE. Because these methods build models based on experimental data, they tend to be far more robust and accurate in their predictions. Unfortunately, because they are built using data from a specific series of compounds or a specific protein receptor, they tend not be very general. LIE and QSAR work well on data similar to that which they were tuned on. [Pg.454]

So why, after 30 years of structure-based drug design method development, is predicting ligand affinity such a difficult and yet to be solved problem We would like to leave you with one last thought.
