Adverse effects include dizziness, postural hypotension, sedation, dry mouth, headache, sleep disturbances, depression, anxiety, impotence, blurred vision, constipation, skin rash, arthralgia, fatigue, anorexia, haemolytic anemia, parkinsonian signs, drug fever and hepatitis. 

Adverse effects include urticaria, fever, rashes, angioedema, anaphylaxis, haemolytic anemia, neutropenia, thrombocytopenia, coagulation disorders, diarrhoea etc.

Several therapeutic agents can promote the development of Thrombotic Micro Angiopathy (TM A), which is characterized clinically by microangiopathic hemolytic anemia, thrombocytopenia, and various ischemic end organ manifestations. Pathologically, vessel wall thickening, endothehal cell swelling, intraluminal platelet thrombi, and microvascular occlusion are noted [158]. Some forms of TMA are dominated by renal impairment and are usually referred to as haemolytic uremic syndrome (HUS) others show predominant central nervous system involvement and are referred to as thrombotic thrombocytopenic purpura (TTP).

Low concentrations of serum inhibit polyene haemolysis [109], so that direct interaction of antibiotic with erythrocytes in vivo may not be the cause of polyene-induced haemolytic anemia observed in patients [353,354]. The survival time of red blood cells in vivo was not reduced by amphotericin B and it was suggested that haemolytic anemia following polyene therapy was the result of suppressed erythrocyte production by bone marrow [353]. 

Prolonged intravenous amphotericin B therapy has been shown to produce haemolytic anemia and renal damage. Although polyene antibiotics are known to lyse erythrocytes in vitro, it is not thought that in vivo lysis caused the observed anemia (see p. 150) but resulted from inhibition of bone marrow erythrocyte synthesis [353].
