Analysis isozyme

Of the thousands of different enzymes present in the human body, those that fulfill functions indispensable to cell vitality are present throughout the body tissues. Other enzymes or isozymes are expressed only in specific cell types, during certain periods of development, or in response to specific physiologic or pathophysiologic changes. Analysis of the presence and distribution of enzymes and isozymes— whose expression is normally tissue-, time-, or circumstance-specific—often aids diagnosis.

Savolainen et ak proposed a division of the angiosperms into two major groups the eudicots (particularly asterids and rosids) and the noneudicots (monocots and Laurales, Magnoliales, Piperales, Ceratophyllales, Amborellaceae, Nymphaceae, Liliaceae). ACS isozymes used in the analysis are presented in the order proposed by molecular systematicians. 

These data agree with the idea that lignifying walls contain specific isozymes, each possessing different affinities towards given substrates. In order to check this hypothesis, an analysis of cell wall peroxidase fractions was undertaken. 

The only unusual feature of the analysis seems to be the high proline content. Also, the values for serine and threonine reported by either Simpson et al. (41) or others (43, 46) are probably reversed. The differences between columns two and three are probably too small to indicate a different composition for the two isozymes.

Willnecker, Jahnke and Buehner96 reported recently the preliminary results of X-ray crystallographic analysis of the potato type-L isozyme at 2.6 A resolution, and showed that the overall folding of the potato phosphorylase is essentially the same as that of the rabbit muscle enzyme (more similar to the 7 -form than to the T-form). Only small differences were found in the core of the subunit, although local structures on the surface including the loops deviated significantly, being consistent with the sequence differences with some minor insertions and deletions as compared with the rabbit muscle enzyme (see above). P. falciparum PNP also proceeds through a TS different than bovine however similar to human isozyme. The TS properties and their interactions in the active site obtained from KIEs analysis provide useful information for inhibitor design.
