Aging of membranes

In factorial designs we have n variable factors which we are able to adjust at fixed levels. Sometimes the factors only exist in discrete levels but sometimes we are interested in or are only able to set up these discrete levels. The assignable causes, the factors which we assume will affect the recorded responses, may be quantitative or qualitative features. The quantitative character of temperature, pH, stirring velocity, etc. is obvious. Examples of qualitative factors are the age of membranes in ion selective electrodes or filter devices (with the levels old and new) or a certain medication or noxious agent (present or absent). In this situation the notion version of a factor makes sense (ISO 3534/3 in [ISO STANDARDS HANDBOOK, 1989]). [Pg.73]

The gene defective in cystic fibrosis codes for CFTR (cystic fibrosis transmembrane condnctance regulator), a membrane protein that pumps CP out of cells. If this CP pump is defective, CP ions remain in cells, which then take up water from the surrounding mucus by osmosis. The mucus thickens and accumulates in various organs, including the lungs, where its presence favors infections such as pneumonia. As noted above, the presence of Met(O) in proteins would go undetected after acid hydrolysis and subsequent amino acid analysis. Thus, since this method of hydrolysis is most commonly used, it is impossible to ascertain from the literature the abundance of Met(O) residues normally present in proteins. However, a number of studies have reported the presence of Met(O) residues in various proteins using one of the appropriate procedures described above. It has been found that Met(O) residues comprise 30% of the total Met in proteins isolated from bovine glomerular basement membranes and anterior lens . Other investigators have reported that the levels of Met(O) in proteins of the trabecular meshwork of human eyes increased with the age of the donor . The amount of Met(O) detected ranged from 15% (10 years old) to 55% (79 years old) of the total methionine content found in the tissue samples. We have already stressed the potential importance of lipid-rich membranes in the skin as potential targets for ROS-induced damage and ageing of human skin is morphologically identical to changes found by peroxidative processes (Serri et al., 1977). The involvement of AA metabolites in skin disease, and in particular psoriasis, has been the subject of much recent interest. Studies have included topical and intradermal administrations of AA metabolites, and assay of such products in clinical specimens. Results show that concentration of AA, 12-hydroxy-eicosatetraenoic acid (12-HETE), PG and leu-kotrienes are increased in psoriatic lesions (Hammarstrom etal., 1975 Camp etal., 1983 Brain etal., 1984 Duell et al., 1988) and also that full-thickness epidermis from normal and diseased skin has the enzymatic capacity to convert AA to some of the same metabolites (Hammarstrom etal., 1975, 1979 Camp etal., 1983 Brain etal., 1984 Ziboh et al., 1984 DueU et al., 1988). Primary and secondary products, and end-products of lipid peroxidation have all been shown to accumulate in senile cataracts (Babizhayev, 1989b Simonelli et al., 1989). Accumulation of these compounds in the lenticular epithelial membranes is a possible cause of damage preceding cataract formation. In senile cataracts there is also extensive oxidation of protein methionine and cysteine in both the membrane and cytosol components (Garner and Spector, 1980), while in aged normal lenses a lesser extent of oxidation was confined to the membrane. Polyion complex technique [40] is a unique method for immobilization of bilayer membranes with polymers. Water-insoluble complex is precipitated as the polyion complex when an aqueous solution of the charged bilayer membrane is mixed with a water solution of the counter charged polyelectrolyte. Stoichiometric ion pair formation is often found.
