The great leap forward for chromatography was the seminal work of Martin and Synge (7) who in 1941 replaced countercurrent liquid-liquid extraction by partition chromatography for the analysis of amino acids from wool. Martin also realized that the mobile phase could be a gas rather than a liquid, and with James first developed (8) gas chromatography (GC) in 1951, following the gas-phase adsorption-chromatographic separations of Phillips (9). [Pg.3]

The primary process of lEC involves adsorption and desorption of ionic species from ionogenic groups located in the packing. lEC was the first of the traditional column techniques to be exploited for modern LC, due principally to the need for the fast routine analysis of amino acids and protein mixtures. [Pg.332]

These methods are of practical importance mainly when closely similar substances have to be separated and therefore the all or none method is insufficient. Some of the results gained, however, are of a rather general interest for the understanding of the processes underlying adsorption analysis and therefore will be dealt with here, before a survey of the other methods of applying adsorption analysis to amino acids are discussed. [Pg.71]

Etzler and Kabat purified the agglutinating principle from Dolichos seed extracts by adsorption to insoluble polyleucyl hog A + H substance.108,517 Specific elution of the adsorbent with 2-acetamido-2-deoxy-D-galactose gave a protein that appeared homogeneous by immunodiffusion, immunoelectrophoresis, disc-gel electrophoresis under acid and alkaline conditions, and sedimentation analysis. The amino acid composition of the protein of molecular weight 141,000 reflected a high content of aspartic acid and serine, little methionine, and no cysteine. Carbohydrate analysis showed a sugar content of 2.4% of hexose, 1.6% of hexosamine, and 1.5% of 2-acetamido-2-deoxy-hexose. [Pg.227]

The colorimetric properties of amino acid terminated PDAs were investigated by diermochromism and solution pH induced chromism. Quantitative analysis of chromatic transition was conducted by analyzing the colorimetric response (CR) as a function of solution pH (IS). The CR is defined as the percent change in the maximum adsorption at 646 nm with respect to the total aWrption at 542 nm and 646 nm. Figure 3 shows the CR vs. pH for the amino acid terminated PDA microstructures. Sigmoidal ciuwes were obtained... [Pg.101]
