Adhesion molecules, monoclonal antibody

Williams et al. (1997) described renal ischemia-reperfusion injury in rats. The animals were anesthetized and subjected to 45min of bilateral renal occlusion using atraumatic vascular clamps before renal perfusion was reestablished. After various time interval (up to lweek) blood urea nitrogen, creatinine and myeloperoxidase activity in the kidney were determined. The protective effects of an intracellular adhesion molecule monoclonal antibody were tested.

Inhibition of inflammatory cytokines (Fig. 2) Humanized monoclonal anti-TNF antibodies (Infliximab (Remicade ), Adalimumab (Humira )) bind with high selectivity to human TNF-a and neutralize its activity. Thereby, infliximab decreases the effects of enhanced TNF levels during inflammatory disease such as production of proteases, chemokines, adhesion molecules, cyclooxygenase products (prostaglandins), and proinflammatory molecules such as interleukin-1 and -6. The antibodies may also recognize membrane-bound TNF-a on lymphocytes and other immune cells. These cells may subsequently become apoptotic or are eliminated via Fc-receptor-mediated phagocytosis.

Abciximab is a Fab fragment of a murine-human monoclonal antibody that binds to the integrin GPIIb/IIIa receptor on activated platelets and inhibits fibrinogen, von Willebrand factor, and other adhesion molecules from binding to activated platelets, thus preventing their aggregation.
