Wheat germ precipitation

White precipitation of proteins from wheat germ extract may be seen when the buffer of the translation solution is exchanged for that with lower pH. Remove the precipitation by centrifugation (20,000g, 15 min). 

The method of lectin precipitation is based on selective precipitation of BALP with wheat germ lectin. The difference between the catalytic concentration before the precipitation and after can relatively successfully quantified in the form of the catalytic concentration of BALP (R7). 

The use of wheat germ lectin, binding the N-acetylglu-cosamine residues present in different amounts on individual isoenzymes, has also been the basis of a commercial technique for the selective precipitation and measurement of bone ALP. However, this technique does not provide sufficient accuracy and precision for quantifying the bone fraction. 

Wheat germ agglutinin (WGA), a lectin, has been used to measure BAP by affinity chromatography, affinity electrophoresis, or differential precipitation of isoforms.

Laboratory studies of protein synthesis usually involve the addition of a radioactively labeled amino acid and either natural or synthetic mRNAs to systems containing the other components. To observe the formation of protein, advantage is taken of the fact that proteins, but not amino acids, can be precipitated by solutions of trichloroacetic acid. Thus, one can observe the extent to which radioactivity has been incorporated into acid-precipitable material as a function of time to estimate the rate of formation of protein. In one such experiment, poly(U) is used as a synthetic mRNA in an in vitro system derived from wheat germ (a eukaryote).

Desulphated keratan sulphate has been shown to interact with wheat-germ agglutinin, supporting the view that non-consecutive (1 -> 4)-2-acetamido-2-deoxy-j8-D-glucopyranosyl residues interact specifically with the lectin to precipitate the complex. The distribution and chemical composition of keratan sulphate in various tissues, including those from a patient with Marfan syndrome, have been related to ageing.

To identify the size of the initial translation product of the 60 kDa biotinylated protein, we isolated RNA from carrot somatic embryos, in vitro translated the mRNA with a rabbit reticulocyte or wheat germ extracts, and immunoprecipitated the said polypeptide. With translation products from the wheat germ extracts the predominant polypeptide that was precipitated was 60 kDa, although a less abundant, 65 kDa polypeptide was also immunoprecipitated. This later polypeptide was the predominant protein immunoprecipitated from the translation products obtained with rabbit reticulocyte extracts (Fig. 2).
