Dissolve (or suspend) 0-25 g. of the acid in 5 ml. of warm water, add a drop or two of phenolphthalein indicator and neutralise carefully with ca. N sodium hydroxide solution. Prepared feeds are marketed in various forms from very fine particles through cmmbles, flakes, and pellets. Pelleted rations may be hard, semimoist, or moist. Hard pellets typically contain less than 10% water and can be stored under cool, dry conditions for at least 90 days without deterioration of quahty. Semimoist pellets are chemically stabilized to protect them from degradation and mold if they are properly stored, while moist pellets must be frozen if they are not used immediately after manufacture. N-Acetylation of Kasugamycinic Acid (9a). A solution of kasugamycinic acid (225 mg.) dissolved in 10 ml. of water was treated with acetic anhydride (0.3 ml.) under cooling sodium bicarbonate was used to keep the pH 7.2 and stirring continued for 30 minutes. The reaction product was passed through Dowex 50W-X2 (H form) and the column was washed with water. The combined filtrate was subjected to lyophilization to afford 234 mg. of a crude N-acetyl derivative. The live fireflies are dried over calcium chloride in a vacuum desiccator, and then their lanterns are separated by hand. An acetone powder prepared from the dried lanterns is extracted with boiling water. The cooled aqueous extract is extracted with ethyl acetate at pH 3.0, and the ethyl acetate layer is concentrated under reduced pressure. Cultures of G. polyedra (L. polyedrum) are grown at 20 5°C in a supplemented sea water medium (Hastings and Sweeney, 1957 Hastings and Dunlap, 1986), under cool-white fluorescent lighting of a 12-hr light/12-hr dark cycle. The cultures are inoculated at densities of 100 to 500 cells/ml. After 2-4 weeks, cells are harvested by vacuum filtration on a filter paper at cell densities of 7,000-15,000 cells/ml, yielding 0.3-0.7 g wet cells per liter of culture. [Pg.251]
