In an interesting aside, many years after these synthetic capsules were first prepared, it was found that cells produce protein vaults that have similar encapsulating abilities but on a much larger scale. Whereas cryptands bind single alkali metal cations, vault proteins bind RNA composed of over 140 bases and other similarly sized guests. These biological crypts are discussed later. [Pg.28]

Finally, a protein has been identified that is unique from the previously discussed ABC proteins. This protein, called lung resistance protein or major vault protein, has a molecular weight of 110 kD and is found not in the cell membrane but rather the nuclear membrane or the cytoplasm. The role of this protein is to prevent the drug from accessing the nucleus by trapping the drug in vesicles called vaults. The vaults then shuttle... [Pg.369]

Figure 18.13. Molecular weight measurements of the human major vault protein (MVP). MALDI-TOF-MS (A) and ESl-QTOF-MS (B) of the denatured MVP protein (pH < 3) show the molecular weight of the MVP chain to be 96.8 kDa, (C) ESI-GEMMA of native MVP (pH 6.8) demonstrates the assembly of the MVP vault to a 96-mer, 9.4-MDa complex.
