Elow rate determines the separation time and can significantly affect resolution and efficiency. The effect of flow rate on HETP for TSK-GEL SW and TSK-GEL SWxi analytical columns is shown in Fig. 4.6. Resolution is typically higher at slower flow rates, although results shown in Fig. 5B indicate that, with increasing sample load, the faster flow rates can give higher resolution. [Pg.101]

TSK-GEL SW columns have also been used to isolate heavy and light chains from immunoglobulins. Kast et al. (7) reported how these chains were desalted and isolated on a 21.5-mm i.d. X 60-cm TSK-GEL G4000SW semipreparative column using a 0.1 trifluoroacetic acid (TEA)/40% acetonitrile mobile phase. [Pg.104]

TSK-GEL PW type columns are commonly used for the separation of synthetic water-soluble polymers because they exhibit a much larger separation range, better linearity of calibration curves, and much lower adsorption effects than TSK-GEL SW columns (10). While TSK-GEL SW columns are suitable for separating monodisperse biopolymers, such as proteins, TSK-GEL PW columns are recommended for separating polydisperse compounds, such as polysaccharides and synthetic polymers. [Pg.106]
