The aim of the present study was to evaluate the effect of LLLT on oxidative markers in serum and tissue biopsies of healing ulcers before and after the 8th session of an LLLT course of chronic leg ulcer treatment. Oxidative damage was assessed in terms of lipid peroxidation reflected by serum malondialdehyde (MDA) level, protein oxidation was measured in terms of tissue protein carbonyls (PCb), and DNA damage was measured in terms of DNA fragmentation. Antioxidative activity was estimated by measuring activity of SOD, GPX and CAT enzymes.

Malignant lesions and normal tissue biopsies (colon) are fixed with formalin and embedded in paraffin (Hernandez-Blazquez et al., 2000). Paraffin sections (4 p,m thick) are deparaffinized, rehydrated, and rinsed in PBS. They are placed in 0.1 mM citrate buffer (pH 3.4), heated for 10 min in a microwave oven at full power (720 W), and washed in PBS. The slides are immersed in 2N HC1 for 2 hr at 37°C and then rinsed in PBS. The sections are covered with 100 p,l ofhybridoma supernatant containing anti-5-MeCyd monoclonal antibody (5 (xg/ml) and incubated for 1 hr at room temperature with a biotinylated goat antimouse secondary antibody diluted 1 200 in PBS containing 0.1% BSA. 

Of the different types of oral mucosal cell cultures that have been used [47,48], the most commonly used ones are explants of primary cultures. Small pieces of excised buccal or sublingual tissue are placed in a support system and fed with culture medium. The outgrowths obtained from these tissue explants are then transferred and grown in appropriate media. For example, outgrowths of fibroblasts [49] thus obtained have been described. Gibbs and Ponec [50] reconstructed the epithelium of mucosal tissue by placing a tissue biopsy (with the epithelial side upwards) onto a fibroblast-populated collagen gel. The explants obtained were cultured immediately at the air liquid interface until the epithelium had expanded over the gel (2-3 weeks). These explant cultures may retain many of the in vivo tissue characteristics.

In the clinic, PK/PD relationships with the direct pharmacological effect, reduction of target mRNA and protein as the response measure, have been demonstrated in two studies with 2 -MOE partially modified ASOs [42, 50]. In a Phase Ila study in patients with rheumatoid arthritis, ISIS 104838 was targeted at the mRNA of TNF-a (a cytokine which is important in immune response and inflammation). The results showed that TNF-a mRNA and protein reduction in synovial tissue biopsies correlated positively with concentrations of ISIS 104838 in the synovial tissues [50]. The demonstration of amyloid protein in a tissue biopsy specimen by staining with Congo red and examination by polarized light microscopy is required for a definitive diagnosis. 

Tissue biopsies were taken from the center of the inner cheek of the oral mucosa in hamster, guinea pig, rat, rabbit, dog and mini-pig. The tissues were fixed in formalin, dehydrated in ethanol and xylene and embedded in parafin wax. The parafin blocks were raicrotomed and the sections stained with Eosin/Haemotoxalin for examination by light microscopy. 

All animals made a full and uneventful recovery from the experimental procedure and no evidence for drug or procedural related adverse reactions was obtained. Mucosal tissue biopsies showed no apparent damage at the microscopic level.
