The Chemical Synthesiser

Oxford chemistry was tripartite in terms of sites. Physical chemistry prospered in two college laboratories, organic chemistry burgeoned in the Dyson Perrins 

Laboratory, and inorganic chemistry was housed in the Old Chemistry Department. If there was a unifying force it was provided by Sidgwick. His synoptic vision, which embraced the main three branches of chemistry, was revealed in his publications, his contributions to colloquia and seminars, and his dominance of both the Alembic Club and the Dyson Perrins tea club. It was rightly said after his death that he was a strict but not severe father of the whole Oxford school of chemistry. 

In 1933 yet another work of synthesis appeared with the publication of his book on the covalent link in chemistry. Like the 1927 book this one took the whole of chemistry for its province. It grew out of his invitation from L. M. Dennis to visit the USA in 1931 as George Fisher Baker lecturer in chemistry at Cornell University for which he was paid 5,000 salary. The optical resolution of the chemically synthesised N-acetyl-DL-phenylalanine by an acylase enzyme is given in reaction 4 (Figure 8.6). A selective hydrolysis of N-acetyl-L-phenylalanine is performed. 

The processes of energisation in cells are, by the nature of the chemicals synthesised, effective in retaining more reduced chemicals than the initial environment, so oxidising equivalents had to be rejected to the environment. The first process was probably the formation of elementary sulfur from H2S. 

Initial sales prices were high, in the order of 4,-8,000 /kg, but as the range of uses, and the volume of sales of T -decalactone increased, sales prices fell to ca 500 /kg. Thus, in just over 10 years, I -decalactone has moved from being very much a specialty chemical to much more of a commodity status with several producers and much bigger volumes of sales. However, even these lower prices are still far higher than for the chemically synthesised I -decalactone. (Gatfield, 1997 Maume and Cheetham, 1991). 

At present most bioprocesses in the organic chemical industry are actually mixed chemical/biochemical processes. In such processes, chemically synthesised educts (chemical precursors) are biotransformed and then re-enter chemical synthesis. The main reason for this approach is that, in general, higher volumetric productivities can be achieved with chemical catalysts. 

Chemically synthesised D,L-hydantoins prepared from the corresponding aldehydes via die Bucherer Berg reaction are converted by the bacterial cells (Bacillus brevis), containing a D-spedfic hydantoinase, to a mixture of D-N-carbamoyl amino acid and L-hydantoin. The latter compound undergoes rapid and spontaneous racemisation under the conditions of the reaction, therefore, in principle 100% of the hydantoin is converted into the D-N-carbamoyl compound. The D-amino add is obtained after treatment of the D-N-carbamoyl compound with nitrous add. This process is operated on an industrial scale by the Japanese firm Kanegafuchi. 

Small interfering RNAs (siRNA) are the mediators of gene-specific silencing by RNA interference. SiRNA stands for small interfering RNA duplexes. They are typically 21-23 bp in length. SiRNA were either chemically synthesised for experimental purposes or produced by Dicer-mediated cleavage of long double-stranded RNA.

The regulation also stipulates that a maximum of three courses of treatment with chemically synthesised allopathic veterinary medical products or antibiotics within one year (or no more than one course of treatment if the productive life cycle is less than one year) is acceptable. These regulations are designed to encourage the use of preventive management and alternative treatments for the control of parasites and diseases. Vaccinations, veterinary medicine treatments for parasites and any compulsory eradication schemes established by Member States are exempt from the treatment maximums, in order to ensure animal welfare. 

The first step in a strategy for developing effective control of a pest by chemical ecology must be to determine if the species uses a chemical signaling system. If intra-specific signaling is shown to occur, the next step is to identify and synthesise the chemical agents utilized, with a view to successful application under field conditions. The most likely source of communication chemicals in amphibians is the secretions of the skin glands.
