Thus, Xi exemplifies both extremes of the histone code, namely the general deacetylation of all four core histones and the specific retention of H3 dimethylated at lysine 9/27. It also illustrates the great stability of epigenetic signals carried by the histone tails. The general underacetylation of Xi, the undermethylation of H3 lysine 4 and the selective retention of H3 dimethylated at lysine 9/27, are all maintained in human x hamster somatic cell hybrids in which Xi is the only human chromosome and therefore exists in an almost completely foreign (i.e., hamster) cellular environment [73].

Somatic cell hybrid The cell resulting from the fusion of animal cells derived from somatic cells that differ genetically. 

Single cell-derived colonies or foci are readily analyzed by cell colony hybridization (Avraham et al., 1989). Although cell colony hybridization on animal cells was described early (Villarreal and Berg, 1977), the subsequently developed dot/slot blot hybridization is normally preferred. Colony hybridization can have some particular advantages, however, such as the quantification of transfected cells or foci, the isolation of certain foci or colonies and may be useful for screening of stable somatic cell hybrids.

The human A locus is located on chromosome 22. As in the case of the human Igh locus, the A genes were first tentatively mapped by studies measuring immunoglobulin gene expression in somatic cell hybrids [91] and the chromosome assignment later confirmed by direct hybridization studies [99].

Omalizumab is a monoclonal antibody developed through somatic cell hybridization techniques and was identified as a murine anti-human IgE antibody, originally called MAE11 (48). It is designed to interact with the site that binds to FceRI on mast cells. Additional amino acid sequences have been incorporated into the antibody so that a humanized product resulted that only differs by 5% nonhuman amino acid residues.
