In the long term, filters and strainers become clogged this is their purpose. Minerals and scale start forming on the internal pipe walls and this reduces the interior diameters on the pipe. A 4 inch pipe will eventually become a 3.5 inch pipe. This moves the pump on its curve beeause as the pipe diameter reduces, the velocity must increase to maintain flow through a smaller orifice. A decrease in plasma volume leads to decreased MAP, which is detected by baroreceptors in the carotid sinuses and the arch of the aorta. Prazosin, oxazosin and terazosin (see p. 73) produce a competitive block of oci adrenoceptors. They decrease peripheral vascular resistance and lower arterial blood pressure by causing the relaxation of both arterial and venous smooth muscle. These drugs cause only minimal changes in cardiac output, renal blood flow, and glomerular filtration rate. Therefore, long-term tachycardia and increased renin release do not occur. Postural hypotension may occur in some individuals. Prazosin is used to treat mild to moderate hypertension and is prescribed in combination with propranolol or a diuretic for additive effects. Reflex tachycardia and first dose syncope are almost universal adverse effects. Concomitant use of a p-blocker may be necessary to blunt the short-term effect of reflex tachycardia. [Pg.200]

Contamination. Unlike the short-term experiments where contamination by other E. coli strains during the course of the experiment is obvious, E. coli contamination is of serious concern for these long-term experiments. Replacement of the culture by the contaminant will appear the same as an adaptive replacement. Strains should be tagged with various genetic markers so they can be distinguished from contaminants. These can be subtle changes such as electrophoretic variants and DNA sequence changes or more obvious ones such as resistance to certain antibiotics. Both pork and human insulin are definitively less immunogenic than beef insulin, producing fewer circulating insulin antibodies, but several studies have indicated no detectable change in antibody concentrations on switching from pork to human insulin or vice versa. Antibodies cause lipoatrophy and are responsible for the substantial insulin resistance seen in some patients, but both events are rare now that purified pork insulin is in common use. Interest has recently been revived in the possible contribution of antibodies in modifying metabolic control. Much controversy seems to exist as to whether the diabetic heart is more or less sensitive to ischemic injury. A large number of studies using animal models of experimental diabetes (induced by streptozotocin or alloxan) report no change, increased or decreased sensitivity to ischemia and reperfusion. Differences in the experimental design in relation to metabolic and ion changes that occur in diabetic heart might be the basis of such discrepancies. Short-term diabetes is associated with decreased sensitivity to zero-flow ischemia and reperfusion injury. This beneficial effect disappears with increased duration or severity of diabetes. The diabetic heart appears to be more vulnerable in low-flow ischemia and in the presence of elevated fatty acids in the perfusate. As mentioned above in section 8.2 and listed in Tables 8.1 and 8.3, heat resistant, organic fibres have chemical structures that are little changed physically or chemically by short-term exposures to temperatures above the 200 and even the 300 °C levels and may be used continuously at temperatures as high as 150 °C. For technical textiles used in high temperature industrial processes, such as gas and liquid filtration, longterm exposures to temperatures of about 100-150 °C are often required.
