Traditionally, Sephadex has been used for molecular weight analyses of proteins (Determan and Brewer, 1975) and clinical dextran (Granath and Kvist, 1967). Today, Sepharose, Sephacryl, or Superose is used for the assay of broad MWDs. Superdex is the premiere choice for attaining the highest resolution of components of similar size (see Fig. 2.1). [Pg.35]

For the purpose of high-resolution fractionation, the gel medium must be tailor made to cope with different separation ranges. The Superdex family is designed for the high resolution of peptides and proteins having a molecular mass of 500 to 100,000. Dextran gels have been utilized since the late 1950s (1) for the separation of biopolymers. First attempts on Sephadex (2-5) and Sephadex/Sepharose (6-8) systems are documented for hydrolyzed and native starch glucans. Up until now, particularly for the preparative and semipreparative separation of polysaccharides, a range of efficient and mechanically stable Sephacryl gels (9-14) have been developped. [Pg.465]

The performance of several Sephacryl gel combinations is illustrated by results achieved for glucans from different types of starch granules. The applied Sephacryl gels of Pharmacia Biotech (15) are cross-linked copolymers of allyl dextran and N,N -methylene bisacrylamide. The hydrophilic matrix minimizes nonspecific adsorption and thus guarantees maximum recovery. Depending on the pore size of the beads, ranging between 25 and 75 im in diameter, aqueous dissolved biopolymers up to particle diameters of 400 nm can be handled. [Pg.465]

Preswelled Sephacryl S-1000 was prepared in a K26/100 column (88 X 2.6 cm). Equilibration with 0.005 M NaOH containing 0.002% NaN3 at a flow rate of 0.67 ml/min was achieved after 20 hr. The mass and iodine-complexing potential of separated glucan components was determined off-line for each of the subsequently eluted 5-ml fractions.
