Screening antimicrobial

Several biologically and pharmacologically active compounds have been prepared from the condensation of the acid chloride of 1-naphthoxyacetic acid with carbazole, iadole, or pyrrole ia 2A[ NaOH solution ia ethanol (63). Also, naphthyloxy derivatives of imidazole, benzimidazole, and benzotriazoles have been synthesized and screened for their antimicrobial, analgesic, and antiinflammatory activities. 2-Naphthyloxy derivatives are comparatively more active than 1-naphthyloxy derivatives (64). 

The major dasses of antibiotics are secondary metabolic products of micro-organisms. Many were discovered by empirically screening culture filtrates or cell extracts for antimicrobial activity. A range of techniques (examples are methods using, impregnated discs, porous cylinders, cut wells, see Figure 6.2) have been used to carry out such screening.

The antimicrobial activity of thiosemicarbazones against Mycobacterium tuberculosis in vitro was first reported by Domagk et al. [28] and later confirmed in vivo [29]. Screening revealed that only certain substituted be-nzaldehyde and heterocyclic thiosemicarbazones possess antitubercular activity [30-32]. The most widely used is p-acetamidobenzaldehyde thiosemicarbazone (trivial name = thiacetazone), 1. 

The glass plate was exposed to microwave irradiation, eluted, and viewed by standard TLC visualization procedures to assess the results of the reaction. In this particular example, the synthesis of an arylpiperazine library (Scheme 4.25) was described, but the simplicity and general utility of the approach for the rapid screening of solvent-free microwave reactions may make this a powerful screening and reaction optimization tool. The extracts of T. affinis, T. arborea, T. divaricata, T. heyrteana, and T. pandacaqui showed some degree of antitumor activity, whereas the extracts of T. malaccensis, T. dichotoma, and T. crassa exhibited hypotensive effects (9). Recently a systematic antimicrobial, antiamoebic, and antiviral screening of 19 different Tabernaemontana species has been carried out with positive results (218).

FSIS has developed a series of overnight, inexpensive, easy to perform swab bioassay tests for screening tissues, body fluids, or feed extracts for antibiotic residues. The swab tests are used on the farm, in the slaughter plant, or in the laboratory for their designated purpose. Swab test results indicate whether antimicrobial activity is present in the sample at or above allowable levels or absent. Further testing with more sophisticated tests is required to identify and quantify the antibiotics producing the antimicrobial activity. These are usually done in a laboratory as required.
