Most detectors are of the differential type, that is their response is proportional to the concentration or mass flow rate of the eluted component. They depend on changes in some physical property of the gas stream, e.g. thermal conductivity, density, flame ionization, electrolytic conductivity, P-ray ionization, in the presence of a sample component. The signal from the detector is fed to a chart recorder, computing integrator or...

Automated dissolution equipment in most cases must be compliant with the FDA electronic records and electronic signatures regulation (21 CFR Part 11). The requirements of the regulation include use of validated systems, secure storage of records, computer generated audit trails, system and data security via limited access privileges, and the use of electronic signatures. 

While validating a production process, several steps were listed as they pertained to each of the components of manufacturing equipment, process conditions, personnel, and so forth. These key elements multiply rapidly when it comes to analytical methods validation. Take, for example, HPLC — the most commonly used method of analysis. A typical analytical method would involve use of columns, pumps, heaters, detectors, controllers, samplers, sensors, recorders, computers, reagents, standards, and operators — put together as a system. Each of these components and systems needs independent validation, followed by a validation of the system. Note that when this equipment is used to manufacture a product such a therapeutic proteins wherein HPLC techniques are used for the purification purpose, then all additional requirements of a manufacturing system also apply, including, but not limited to, the requirement that the equipment be of a sanitary kind. This limits the choice for manufacturers, and these considerations should be taken into account in the first selection of equipment. 

The reporting gas chromatograph consolidates all the functions of the gas chromatograph, strip chart recorder, computer automation, and computing and reporting integrator into one unit providing a powerful and cost effective solution in both routine and research applications.

When a sample is injected, the injector port is at a temperature sufficient to vaporize the sample components. Based on the solubility and volatility of these components with respect to the stationary phase, the components separate and are swept through the column by the carrier gas to a detector, which responds to the concentration of each component. The detector might not respond to all components. The electronic signal produced as the component passes through the detector is amplified by the electrometer, and the resulting signal is sent to a recorder, computer, or electronic data-collecting device for quantitation. 

In order to remedy problems quickly or to avoid system errors, appropriate procedural controls must be applied to (a) enable early identification of an imminent breakdown, and (b) initiate appropriate preventive action. The next section suggests a course of action to follow to identify, analyze, and record computer system incidents, and to implement and test the corrective action taken.

In this way, X2 serves as a reference measurement for the observed absorbance change to be studied at Xj.
