Purple spots

Symptoms Attacks young growth. Purple spots are found on canes, leaves, blossoms, and stalks. Leaves may drop, bark can split, and small cankers form. Fruit yield is reduced. Severe infection will cause distortion and death. 

The eluent front was marked on the glass plates prior to drying over a hot plate. Amino acids were visualized as purple spots by application of the ninhydrin spray reagent to the plate and drying with hot air. Amino acids with zero mobility (Rf = 0 in BUOH/HAC/H2O) are assumed to be cross-links (Keller et al., 1984).

Spray the paper chromatogram lightly with ninhydrin reagent and heat for 5 min in a 100°C oven. The amino acids will appear as blue or purple spots (except proline, which will appear yellow). Figure 6-9 shows the reaction that amino acids undergo with ninhydrin.

Leaves with spots. Cause Tuliptree spot gall. Circular brown or purple spots surrounded by a circle of yellow on the leaves reveals the presence of tuliptree spot gall, caused by a kind of midge. Damaged leaves may fall early. While unsightly, the problem is not serious and is best countered by removing all affected fallen leaves.
