The anti-inflammatory agent Oxapro2in, 2-(4,5-diphenyl-2-oxa2ole)propionic acid mono aluminum and dihydroxyalurninum salts, is made by reaction of the sodium salt with aluminum sulfate under controlled conditions (96). Again, the aluminum salts of many carboxylic acid based dmgs are less irritating, ulcerous, and/or toxic, and have a more pleasant taste than their parent acids. 

Somewhat milder oxidative conditions lead to loss of but one carbon. Periodic acid cleavage of the side chain in 65, leads to the so-called etio acid (66). Reaction with propionic anhydride leads to acylation of the 17-hydroxyl group (67). Possibilities for neighboring group participation severely limit the methods available for activating the acid for esterification. Best results seemed to have been obtained by use of a mixed anhydride from treatment with diphenyl chloro-... 

A mixture of 50 grams of a-dl-1,2-diphenyl-2-hydroxy-3-methyl-4-dimethylaminobutane hydrochloride, 50 grams of propionic anhydride and 50 cc of pyridine was refluxed for about 5 hours. The reaction mixture was cooled to 50°C and ethyl ether was added to the point of incipient precipitation. The hydrochloride salt of 0 -dl-l,2-diphenyl-2-propion-oxy-3-methyl-4-dimethylamlnobutane formed in the reaction precipitated upon cooling and was removed by filtration and washed with anhydrous ether. On recrystallization from a mixture of methanol and ethyl acetate, a-dl-l, 2-diphenyl-2-propionoxy-3-methyl-4-dimethyl amlnobutane hydrochloride melted at 170°-171°C.

BODIPY 530/550 C3-SE is 4,4-difluoro-5,7-diphenyl-4-bora-3a,4a-diaza-S-indacene-3-propionic add, sucdnimidyl ester (Molecular Probes). The compound is an analog of BODIPY 530/550 C3, which contains an active NHS ester on its propionic acid side chain (Chapter 2, Section 1.4). The ester reacts with primary amines and other nucleophiles to form stable amide bonds. 

The reaction of succinic acid anhydride 111 with 2-methyl-2,4-diphenyl-2,3-dihydroli/-benzo[Z ][l,4]diazepine 112 in toluene in the presence of potassium carbonate leads to 4-(2-methyl-2,4-diphenyl-2,5-dihydro-l/f-l,5-benzodiazepin-5-yl)-4-oxobutanoic acid 113 in 70% yield [110], while its treatment with 2,4-diphenyl-2,3-dihydro-l//-l,5-benzodiazepine 104 follows with chalcone elimination and yields 3-(benzimidazol-2-yl)propionic acid 114 [115] (Scheme 4.36).

Ethyl acetate and ethyl propionate in ethanolic sodium ethoxide with 5-aminoimidazole-4-carboxamide and its riboside have provided a route to 2-methyl- and 2-ethyl-hypoxanthine arid corresponding 2-alkylinosines, respectively (67JOC3258,68JA2661). 2-Methylhypoxan-thine has also been prepared from the same imidazole and ethyl orthoacetate via an intermediate ethoxymethylene derivative which cyclized to the purine when heated (60JA3144). The same method has been applied to the synthesis of 2,8-dimethyl- and XS-diphenyl-hypoxanthine from appropriate imidazole precursors. 

Thallium diphenyl acid propionate forms fine silky needles from toluene, M.pt. 164° C., these being completely soluble in cold chloroform, hot alcohol, pyridine, ethyl acetate, or glacial acetic acid, fairly soluble in boiling water, and insoluble in ether or light petroleum. 

Thallium diphenyl acid valerate.— M.pt. 176° Cv forms fine white needles from toluene, having a similar solubility to the propionate.
