One mol of a-bromopropio-ortho-toluidide is mixed with a solution of 3 mols of n-propyl-amine in 500 ml of water-free benzene and the reaction mixture is heated in an autoclave to 80°C for 8 hours. After cooling the reaction mixture is treated as described above. The base is obtained as a colorless oil. BP 159 162°C/0.1 mm. Yield 55%. The base is then converted to the hydrochloride by reaction with HCI. [Pg.1294]


Prilocaine hydrochloride [1786-81-8] is also similar in profile to Hdocaine, although prilocaine has significantly less vasodilator activity. Prilocaine is the least toxic of the amino amide local anesthetics. However, its tendency to cause methemoglobinemia, especially in newborns, has eliminated its use in obstetric surgery. [Pg.415]

It is a local anaesthetic of the amide type which is employed for surface, infiltration and nerve block anaesthesia. Its duration of action is in between the shorter-acting lidocaine and longer-acting mepivacaine. It possesses less vaso-dilator activity than lidocaine and hence may be used without adrenaline. Therefore, solutions of prilocaine hydrochloride are specifically beneficial for such patients who cannot tolerate vasopressor agents patients having cardiovascular disorders, diabetes, hypertension and thyrotoxicosis. [Pg.145]
